Research Objectives
Laboratory animal models used in basic and biomedical research are essential to understanding human health and disease and facilitating the development of diagnostic approaches and therapeutic interventions. A large number of animal models, including Drosophila and zebrafish, are being generated at an unprecedented pace due to the rapid advancements in genetic tools, particularly gene editing technology. However, this rapid increase in genetic stocks is creating challenges in maintenance, preservation and sustainability of these resources in reliable and cost-effective ways. Cryogenic or other long-term preservation and revival of such genetic stocks will be key in ensuring their cost effective and reliable maintenance and distribution to support biomedical research.
The fruit fly (Drosophila melanogaster) and the zebrafish (Danio rerio) are two of the leading non-mammalian model organisms used for understanding the basic biology of animal systems and for modeling human health and disease because of their small size, relatively short generation time, easy manipulation and optical transparency of embryos as well as genetic tractability. Both are important for studies in diverse scientific areas, including genetics, pharmacology, toxicology, neurobiology, behavior and developmental biology. However, cryogenic or other long-term preservation and revival approaches in these species have not been fully established and, in many cases, have proven to be unsuccessful. Currently there are over 60,000 Drosophila stocks at the NIH-supported Bloomington Drosophila Stock Center and nearly 11,000 fish lines at the Zebrafish International Resource Center. These numbers are increasing at a rapid pace. Drosophila stocks are maintained as labor intensive live cultures, while zebrafish stocks are kept as live cultures or cryopreserved as sperm. Therefore, there is an urgent need to achieve simple, reliable, reproducible and cost-effective methods for cryogenic or other long-term preservation and revival of Drosophila and zebrafish stocks, where preservation options are essentially absent (fruit fly) or limited (zebrafish) as compared to the cryopreservation methods developed and widely used for several other animal models, such as worms, mice and rats.
The Office of Research Infrastructure Programs (ORIP) convened a workshop in 2016 on Cryopreservation of Drosophila Strains and in 2017 on Cryopreservation of Aquatic Biomedical Models to offer forums for discussion of major scientific developments and roadblocks that prevent cryogenic or other long-term preservation and revival of Drosophila at any stage of development or zebrafish (with the exception of sperm). Taking into consideration the recommendations of participants at both workshops, ORIP encourages applications under the Exploratory/Developmental Research R21 grant mechanism aimed at developing and improving technologies for cryogenic or other long-term preservation and revival of either Drosophila or zebrafish genetic stocks. Examples of applications include, but are not limited to, the following for cryogenic or other long-term preservation and revival of any developmental or adult forms of Drosophila, including sperm, oocytes, fertilized eggs, embryos, larvae and adult flies, and embryos of zebrafish:
A. Development of germplasm cryogenic or other long-term preservation procedures and tools to achieve efficient, reliable, reproducible and cost-effective approaches for genetic banking and revival, including vitrification methods;
B. Specialized reagents to minimize cellular damage to a wide variety of preserved stocks during collection, processing, storage and revival. Examples include developing reagents for vitrification (Drosophila or zebrafish) or drying (Drosophila);
C. Methods to extend lifespan and reproductive lifespan that can reduce stock maintenance effort, including transgenic and dietary approaches (Drosophila);
D. Novel methods for dehydration of implantable gonad tissues (Drosophila);
E. Tools and processes to standardize cryogenic or other long-term preservation protocols that can boost the reliability, reproducibility, simplicity, throughput capacity and efficiency of stock preservation of either species;
F. Biosecurity methods or protocols for the identification and elimination of pathogens or opportunistic pathogenic microorganisms when preserved resources are revived;
G. High-throughput technologies, including microfluidic technology, that can improve the efficiency of the cryogenic or long-term preservation and revival of stocks;
H. Identification of biological markers for evaluating the status and efficiency of preservation and revival of stocks.
Preference will be given to research projects that are aimed at developing cost-effective and reliable approaches to cryogenic or other long-term preservation and revival of Drosophila or aquatic models, especially zebrafish stocks. Priority will be given to approaches that can be widely used by individual laboratories and stock centers. For zebrafish, preference will be further given to applications that aim to increase the cryopreservation efficiency of embryos. Submissions are acceptable for developing methods that can extend the health and reproductive capability of live cultures and hence significantly reduce the turn-over frequency of live cultures and maintenance effort. The application should include a method to revive preserved stocks, although a high revival rate is not required for this exploratory FOA. The application should clearly describe the characteristics of the technology and the milestones of technology development.
While ORIP is interested in any application that fits within the scope of this FOA, the National Institute of General Medical Sciences (NIGMS) is interested in concepts or prototype technology to enhance embryo preservation or that advance mechanistic knowledge about the cellular and systemic response to preservation conditions, within the mission of NIGMS. Projects that focus on later stage organogenesis or that have a specific disease focus would not be within the scope of the mission of NIGMS.

Only the review criteria described below will be considered in the review process. Applications submitted to the NIH in support of the NIH mission are evaluated for scientific and technical merit through the NIH peer review system.
The R21 exploratory/developmental grant supports investigation of novel scientific ideas or new model systems, tools, or technologies that have the potential for significant impact on biomedical or biobehavioral research. An R21 grant application need not have extensive background material or preliminary information. Accordingly, reviewers will emphasize the conceptual framework, the level of innovation, and the potential to significantly advance our knowledge or understanding. Appropriate justification for the proposed work can be provided through literature citations, data from other sources, or, when available, from investigator-generated data. Preliminary data are not required for R21 applications; however, they may be included if available.
Does the project address an important problem or a critical barrier to progress in the field? Is the prior research that serves as the key support for the proposed project rigorous? If the aims of the project are achieved, how will scientific knowledge, technical capability, and/or clinical practice be improved? How will successful completion of the aims change the concepts, methods, technologies, treatments, services, or preventative interventions that drive this field?
Does the application adequately address the critical knowledge and technology gaps in developing cryogenic or other long-term preservation and revival methods for Drosophila or zebrafish genetic stocks? Does the application sufficiently describe the impact of the proposed technology on stock preservation and revival in large stock centers and/or individual laboratories?
Are the PD(s)/PI(s), collaborators, and other researchers well suited to the project? If Early Stage Investigators or those in the early stages of independent careers, do they have appropriate experience and training? If established, have they demonstrated an ongoing record of accomplishments that have advanced their field(s)? If the project is collaborative or multi-PD/PI, do the investigators have complementary and integrated expertise; are their leadership approach, governance and organizational structure appropriate for the project?
Does the application challenge and seek to shift current research or clinical practice paradigms by utilizing novel theoretical concepts, approaches or methodologies, instrumentation, or interventions? Are the concepts, approaches or methodologies, instrumentation, or interventions novel to one field of research or novel in a broad sense? Is a refinement, improvement, or new application of theoretical concepts, approaches or methodologies, instrumentation, or interventions proposed?
Are the overall strategy, methodology, and analyses well-reasoned and appropriate to accomplish the specific aims of the project? Have the investigators included plans to address weaknesses in the rigor of prior research that serves as the key support for the proposed project ? Have the investigators presented strategies to ensure a robust and unbiased approach, as appropriate for the work proposed? Are potential problems, alternative strategies, and benchmarks for success presented? If the project is in the early stages of development, will the strategy establish feasibility and will particularly risky aspects be managed? Have the investigators presented adequate plans to address relevant biological variables, such as sex, for studies in vertebrate animals or human subjects?
Does the application adequately describe the characteristics of the technology and the feasibility of developing easily applied cryogenic or other long-term preservation and revival technology for Drosophila or zebrafish stocks?
Has the investigator proposed effective methods to determine the reliability of the proposed technology for preservation and revival of a sufficient number of stocks of different genetic backgrounds?
Are any reliable methods proposed to assess the applicability of the proposed technology for implementation in large stock centers and/or individual laboratories?
Does the application describe the ability and technical merits of the proposed technology for preserving a sufficient number of materials, such as embryos, for effective revival of stocks?
Does the application include clear and achievable milestones in terms of long-term viability and revival of preserved materials?If the project involves human subjects and/or NIH-defined clinical research, are the plans to address 1) the protection of human subjects from research risks, and 2) inclusion (or exclusion) of individuals on the basis of sex/gender, race, and ethnicity, as well as the inclusion or exclusion of individuals of all ages (including children and older adults), justified in terms of the scientific goals and research strategy proposed?
Will the scientific environment in which the work will be done contribute to the probability of success? For research that involves human subjects but does not involve one of the categories of research that are exempt under 45 CFR Part 46, the committee will evaluate the justification for involvement of human subjects and the proposed protections from research risk relating to their participation according to the following five review criteria: 1) risk to subjects, 2) adequacy of protection against risks, 3) potential benefits to the subjects and others, 4) importance of the knowledge to be gained, and 5) data and safety monitoring for clinical trials.
For research that involves human subjects and meets the criteria for one or more of the categories of research that are exempt under 45 CFR Part 46, the committee will evaluate: 1) the justification for the exemption, 2) human subjects involvement and characteristics, and 3) sources of materials. When the proposed project involves human subjects and/or NIH-defined clinical research, the committee will evaluate the proposed plans for the inclusion (or exclusion) of individuals on the basis of sex/gender, race, and ethnicity, as well as the inclusion (or exclusion) of individuals of all ages (including children and older adults) to determine if it is justified in terms of the scientific goals and research strategy proposed. The committee will evaluate the involvement of live vertebrate animals as part of the scientific assessment according to the following criteria: (1) description of proposed procedures involving animals, including species, strains, ages, sex, and total number to be used; (2) justifications for the use of animals versus alternative models and for the appropriateness of the species proposed; (3) interventions to minimize discomfort, distress, pain and injury; and (4) justification for euthanasia method if NOT consistent with the AVMA Guidelines for the Euthanasia of Animals. Reviewers will assess the use of chimpanzees as they would any other application proposing the use of vertebrate animals.
