Section I. Funding Opportunity Description
1. Research Objectives
Purpose
This Funding Opportunity Announcement (FOA), issued by the National Institute of Mental Health (NIMH), invites Phased Innovation (R21/R33) grant applications from organizations /institutions that propose the development and characterization of novel mouse models that express human genes or human genetic elements that can aid in understanding the molecular mechanisms underlying brain development and function relevant to candidate genes/gene dosing abnormalities implicated in bipolar disorder, schizophrenia, depression, anxiety, and developmental disorders (http://www.nimh.nih.gov/healthinformation/index.cfm).
Background
In recent years, a number of genes have emerged as candidates for involvement in human mental disorders. Whole-genome association studies continue to identify potentially important genetic factors that may play a role in the etiology of psychopathologies. Although currently unclear, some of these may be specific disease alleles with polymorphisms present either in coding regions or in regulatory elements altering the function or expression characteristics of particular genes. Other genes may, in isolation or in combination, be protective for unknown functional reasons. In addition, there are indications that gene dosage might play a role in various disorders of interest to NIMH. At present, little is known about the mechanisms or pathways that contribute to functional changes for these human gene products or the consequences of these changes on brain function or behavior. One approach to studying candidate genes is to examine the functional impact of variations in intact systems. Expressing specific human alleles of interest in mice may help elucidate gene/protein regulation and function and can provide novel model systems to study human genes related to brain development, physiology, pharmacology, neurochemistry, and behavior. While this approach is clearly exploratory, the mouse model represents a highly tractable system in which to examine the function of genes and genetic variants. The approach would provide critical information regarding the functional significance of human genes/proteins and has already been used with some success in multiple systems, including in the context of neurological conditions.
Nature of the Research Opportunity
This opportunity will stimulate the creation and validation of novel mouse models expressing human candidate genes/genetic elements that have the potential to inform understanding of brain and behavioral function relevant to the etiology (e.g., candidates that confer risk, susceptibility, resilience) or treatment of mental disorders. The FOA uses the NIH Exploratory/ Developmental Grant (R21) and the NIH Exploratory/ Developmental Grant Phase II Phased Innovation (R33) award mechanisms. The NIH envisions that the primary activities of the R21 phase will include making constructs, creating/breeding the genetically altered mice, and initial functional characterization of the mouse line(s). Upon achieving agreed-upon milestones for the R21 phase, investigators may be eligible to receive one additional year of support in the R33 phase to continue characterization and evaluation of mice. This FOA will only support studies to generate and characterize new mouse models beginning with the R21 phase. Applications proposing to study existing mouse lines, even those lacking characterization data will not be considered for support through this FOA.
In general, it is hoped that knowledge will be gained regarding functional aspects of human genes/genetic elements/gene products with regard to brain function and/or behavior relevant to NIMH research priorities. The following objectives would make appropriate topics for proposed R21/R33 projects. This list is not meant to be all-inclusive, but can provide guidance on the types of projects that are of interest to NIMH.
Types of research and experimental approaches that are being
sought to achieve the objectives: A number of approaches to create mice
expressing human genes may be used to achieve the objectives outlined in this
announcement. The approach should be well-justified and take into consideration expression
of the
endogenous mouse ortholog(s) as well as issues related to
regulation of expression. Novel strategies such as recombinase-mediated
cassette exchange (RMCE), recombinase-mediated genomic replacement (RMGR) may be used. Conventional gene targeting
strategies, transgenics, or alternate approaches may
be appropriate as long as investigator justifies the approach and has considered alternatives. Genetic background of the mouse lines should be carefully considered and
justified based on the nature of the genetic manipulation, proposed mouse characterization protocol(s), and/or other issues. Proposed evaluation of the mice in the latter part of the
R21 phase and throughout the R33 phase will be specific to important functional
questions related to the human gene/genetic element expressed. Evaluation and characterization of the resulting mice may include gene/protein expression characteristics, biochemical,
physiological, morphological, and/or behavioral measures or other specific
validations.

II. Award Information
1.
Mechanism of Support
This Funding Opportunity Announcement (FOA)
will use the NIH Exploratory/Developmental Grant and Exploratory/Developmental
Grant Phase II Phased Innovation Grant (R21/R33) award mechanism. The
applicant will be solely responsible for planning, directing, and executing the
proposed project.
This FOA uses Just-in-Time information concepts. For both the R21 and the R33 phases, applicants must complete and submit detailed budget requests using the SF424 Research and Related (R&R) Budget component provided in the SF424 (R&R) Application Package and the SF424 (R&R) Application Guide (see specifically Section 4.7, R&R Budget Component, of the Application Guide). Modular budgets are not permitted for this funding opportunity.
Exploratory/developmental grant support is for new projects only; competing renewal (formerly competing continuation ) applications will not be accepted.
2.
Funds Available
The NIMH intends to commit approximately $1,750,000 dollars in FY08 to fund 7-10 applications. The total project period for an application submitted in
response to this funding opportunity may not exceed 3 years. For the R21 phase
of the application, direct costs are limited to $200,000 over a two-year
period, with no more than $150,000 in direct costs allowed in any single year. The
R33 phase of the application is one year and direct costs must be limited to no
more than $150.000.
Because the nature and scope of the proposed research will vary from application to application, it is anticipated that the size and duration of each award will also vary. Although the financial plans of the Institutes and Centers (ICs) provide support for this program, awards pursuant to this funding opportunity are contingent upon the availability of funds and the submission of a sufficient number of meritorious applications.
NIH grants policies as described in the NIH Grants Policy Statement will apply to the applications submitted and awards made in response to this FOA.
Facilities and Administrative (F&A) costs requested by consortium participants are not included in the direct cost limitation. See NOT-OD-05-004, November 2, 2004.
The award of R33
funds will be based on program priorities, on the availability of funds, and on
the successful completion of scientific milestones as determined by NIH staff
in the context of peer review recommendations.

Any individual with the skills, knowledge, and resources necessary to carry out the proposed research as the Project Director/Principal Investigator (PD/PI) is invited to work with his/her organization to develop an application for support. Individuals from underrepresented racial and ethnic groups as well as individuals with disabilities are always encouraged to apply for NIH support.
More than one PD/PI, or multiple PDs/PIs, may be designated on the application for projects that require a team science approach that clearly does not fit the single-PD/PI model. SPECIAL INSTRUCTIONS
Applications with Multiple PDs/PIs
When multiple PDs/PIs are proposed, NIH requires one PD/PI to be designated as the "Contact PI, who will be responsible for all communication between the PDs/PIs and the NIH, for assembling the application materials outlined below, and for coordinating progress reports for the project. The contact PD/PI must meet all eligibility requirements for PD/PI status in the same way as other PDs/PIs, but has no other special roles or responsibilities within the project team beyond those mentioned above.
Information for the Contact PD/PI should be entered in item 15 of the SF424(R&R) Cover component. All other PDs/PIs should be listed in the Research & Related Senior/Key Person component and assigned the project role of PD/PI. Please remember that all PDs/PIs must be registered in the eRA Commons prior to application submission. The Commons ID of each PD/PI must be included in the Credential field of the Research & Related Senior/Key Person component. Failure to include this data field will cause the application to be rejected.
All projects proposing Multiple PDs/PIs will be required to include a new section describing the leadership of the project.
Multiple PD/PI Leadership Plan: For applications designating multiple PDs/PIs, a new section of the research plan, entitled Multiple PD/PI Leadership Plan (Section 14 of the Research Plan Component in the SF424 (R&R)), must be included. A rationale for choosing a multiple PD/PI approach should be described. The governance and organizational structure of the leadership team and the research project should be described, including communication plans, process for making decisions on scientific direction, and procedures for resolving conflicts. The roles and administrative, technical, and scientific responsibilities for the project or program should be delineated for the PDs/PIs and other collaborators.
If budget allocation is planned, the distribution of resources to specific components of the project or the individual PDs/PIs should be delineated in the Leadership Plan. In the event of an award, the requested allocations may be reflected in a footnote on the Notice of Award.
Applications Involving a Single Institution
When all PDs/PIs are within a single institution, follow the instructions contained in the SF424 (R&R) Application Guide.
Applications Involving Multiple Institutions
When multiple institutions are involved, one institution must be designated as the prime institution and funding for the other institution(s) must be requested via a subcontract to be administered by the prime institution. When submitting a detailed budget, the prime institution should submit its budget using the Research & Related Budget component. All other institutions should have their individual budgets attached separately to the Research & Related Subaward Budget Attachment(s) Form. 3.B. Submitting an Application Electronically to the NIH
To submit an application in response to this FOA, applicants should access this

The NIH will not accept any application in response to this funding opportunity that is essentially the same as one currently pending initial review, unless the applicant withdraws the pending application. 5. Funding Restrictions
All NIH awards are subject to the terms and
conditions, cost principles, and other considerations described in the NIH Grants
Policy Statement.
Pre-award costs are allowable. A grantee may, at
its own risk and without NIH prior approval, incur obligations and expenditures
to cover costs up to 90 days before the beginning date of the initial budget
period of a new award if such costs: are necessary to conduct the project, and
would be allowable under the grant, if awarded, without NIH prior approval. If
specific expenditures would otherwise require prior approval, the grantee must
obtain NIH approval before incurring the cost. NIH prior approval is required
for any costs to be incurred more than 90 days before the beginning date of the
initial budget period of a new award.
The incurrence of pre-award costs in anticipation of a
competing or non-competing award imposes no obligation on NIH either to make
the award or to increase the amount of the approved budget if an award is made
for less than the amount anticipated and is inadequate to cover the pre-award
costs incurred. NIH expects the grantee to be fully aware that pre-award costs
result in borrowing against future support and that such borrowing must not
impair the grantee's ability to accomplish the project objectives in the
approved time frame or in any way adversely affect the conduct of the project.

Milestones:
A description of milestones to be achieved in the R21 phase is required. The milestones proposed in the application should be well-described, quantifiable, and scientifically justified to allow program staff to assess progress in the R21 phase. The milestones will be considered in evaluating the approach proposed by the investigator. A discussion of the milestones relative to the progress of the R21 phase and the implications of successful completion of the milestones for the R33 phase should be included. Applications lacking this information, as determined by the NIH staff will be returned to the applicant without review. The clarity and completeness of the R21/R33 application with regard to specific goals and feasibility milestones are critical. The proposed milestones should not be more than one page and are to be included within the 25-page limit.
Prior to funding an application, the Program Officer will contact the applicant to discuss the proposed milestones and any changes suggested by the review panel as indicated in the Summary Statement. The Program Officer and the applicant will negotiate and agree on a final set of milestones. These will be the basis for judging the success of the R21 work. For funded applications, the Project Director/Principal Investigator (PD/PI) will submit a progress report to the Program Officer upon completion of the R21 milestones. This progress report will be subject to an administrative program review to determine whether to award the R33 component. The release of R33 funds will be based on successful completion of negotiated scientific milestones, on the program priorities, on the predicted impact of additional studies, and on the availability of funds.
The R21 and R33 cannot be funded in the same fiscal year.
1. In the Research and Related Budget component Section K ( Budget Justification )
For the R21 phase of the application, direct costs are limited to a maximum of $200,000 over two years with no more than $150,000 requested in any single year. For the R33 phase of the application, direct costs are limited to no more than $150,000 per year. R21 budgets can only exceed this cap to accommodate Facilities and Administrative (F&A) costs of subcontracts to the project. The combined R21/R33 application is limited to three years in duration with the R21 phase occurring in years 1 and 2 and the R33 phase (if awarded) occurring in year 3.
The application should provide a detailed budget for each Budget Period of the R21 phase and the R33 phase. All budgets should include a written justification Section K, Budget Justification. In Section K of each Budget Period, the application should indicate whether it is the R21 or the R33 phase. The modular budget format is not permitted for this funding opportunity.
2. In the PHS398 Research Plan Component Research Plan Attachments:
Item 1: Specific Aims.
A specific section labeled Milestones should appear at the beginning of the Specific Aims attachment (see requirements for the Milestones section above). The advantage of the combined R21/R33 mechanism is that it offers a seamless transition between the exploratory phase and the development phase of a project. Milestones should be well described, quantifiable, and scientifically justified. Applications should write the milestones to allow for assessment of progress at the end of the R21 phase. Milestones should not be simply a restatement of the specific aims or a timeline. Applications lacking this information will be returned to the applicant without review.
Include sections titled R21 Phase Specific Aims and R33 Phase Specific Aims . For each section, state the specific objectives of the research effort of that phase. Since the goal of the R21 phase of the FOA is the development and initial characterization of novel mouse models containing human genes/genetic elements, hypothesis testing per se, may not be the driving force in developing the proposed research and therefore may not be applicable. For the R21 portion of the grant application, preliminary data are not required, although they should be included when available.
Item 2: Background and Significance.
Explain the scientific rationale for developing novel mice expressing the proposed human gene or genes. Elaborate on the innovative nature of the proposed research and likely impact on biomedical research. Clarify how the fundamental tools and data to be developed are likely to result in significant improvement over existing approaches. Discuss limitations or caveats that may preclude success.
Item 3: Preliminary Studies.
While preliminary data are not required for submission of the R21 phase, this section should provide the current thinking or evidence in the field that substantiates the feasibility of the R21 and R33 phases. If the applicant does have preliminary data, it should be presented in this section.
Item 4: Research Design and Methods.
Follow the instructions in the SF424 Application Guide. Include headers titled R21 Phase Research Design and Methods and R33 Phase Research Design and Methods, and address the research design and methods for the R21 and R33 phase in the appropriate subsection.
Plan for Sharing Research Data
The precise content of the data-sharing plan will vary, depending on the data
being collected and how the investigator is planning to share the data.
Applicants who are planning to share data may wish to describe briefly the
expected schedule for data sharing, the format of the final dataset, the
documentation to be provided, whether or not any analytic tools also will be
provided, whether or not a data-sharing agreement will be required and, if so,
a brief description of such an agreement (including the criteria for deciding
who can receive the data and whether or not any conditions will be placed on
their use), and the mode of data sharing (e.g., under their own auspices by
mailing a disk or posting data on their institutional or personal website,
through a data archive or enclave). Investigators choosing to share under their
own auspices may wish to enter into a data-sharing agreement. References to
data sharing may also be appropriate in other sections of the application.

The NIH R21 exploratory/developmental grant is a mechanism for supporting novel scientific ideas or new model systems, tools, or technologies that have the potential to significantly advance our knowledge or the status of health-related research. Because the Research Plan component is limited to 15 pages, an exploratory/developmental grant application need not have extensive background material or preliminary information as one might normally expect in an R01 application. Accordingly, reviewers will focus their evaluation on the conceptual framework, the level of innovation, and the potential to significantly advance our knowledge or understanding. Reviewers will place less emphasis on methodological details and certain indicators traditionally used in evaluating the scientific merit of R01 applications, including supportive preliminary data. Appropriate justification for the proposed work can be provided through literature citations, data from other sources, or, when available, from investigator-generated data. Preliminary data are not required for R21 applications; however, they may be included if available.
The goals of NIH supported research are to advance our understanding of biological systems, to improve the control of disease, and to enhance health. In their written critiques, reviewers will be asked to comment on each of the following criteria in order to judge the likelihood that the proposed research will have a substantial impact on the pursuit of these goals. Each of these criteria will be addressed and considered in assigning the overall score, weighting them as appropriate for each application.
Note that an application does not need to be strong in
all categories to be judged likely to have major scientific impact and thus
deserve a high priority score. For example, an investigator may propose to
carry out important work that by its nature is not innovative but is essential
to move a field forward.
Significance: Does this study address an important problem? If the aims of the application
are achieved, how will scientific knowledge or clinical practice be advanced?
What will be the effect of these studies on the concepts, methods,
technologies, treatments, services, or preventative interventions that drive
this field? Are the human
genes/genetic elements to be expressed in mice of significance in terms of advancing the research priorities of NIMH? Do the proposed candidate genes/genetic elements have the potential to inform understanding of brain and
behavioral function relevant to the etiology (e.g., candidates that confer
risk, susceptibility, resilience) or treatment of mental disorders.
Approach: Are the conceptual or
clinical framework, design, methods, and analyses adequately developed, well
integrated, well reasoned, and appropriate to the aims of the project? Does the
applicant acknowledge potential problem areas and consider alternative tactics?
Applicants should discuss potential pitfalls in both the mouse generation and
validation stages. For
applications designating multiple PDs/PIs, does the Leadership Plan ensure that
there will be sufficient coordination and
communication among the PDs/PIs? Are the administrative plans for the
management of the research project appropriate, including plans for resolving
conflicts?
Innovation: Is the project original and innovative? For example: Does
the project challenge existing paradigms or clinical practice; address an
innovative hypothesis or critical barrier to progress in the field? Does the
project develop or employ novel concepts, approaches, methodologies, tools, or
technologies for this area?
Investigators: Are the PD/PI(s) and other key personnel appropriately trained and well suited to
carry out this work? Is the work proposed appropriate to the experience level
of the principal investigator and other researchers? Does the PD/PI(s) and investigative
team bring complementary and integrated expertise to the project (if
applicable)?
Environment: Do(es) the scientific environment(s) in which the work will
be done contribute to the probability of success? Do the proposed studies
benefit from unique features of the scientific environment, or subject
populations, or employ useful collaborative arrangements? Is there evidence of
institutional support?
Milestones:
Are the milestones quantitative? Are the proposed milestones appropriate for judging the success of the R21 work? Are the proposed milestones appropriate for judging the potential impact of the R33 phase? Are the proposed milestones appropriate in determining whether the R33 phase should be awarded? Do the milestones establish feasibility for all aspects of the proposed work?
For the R21/R33 Phased Innovation Award application, the initial review group will evaluate the specific goals for each phase and the feasibility milestones that would justify expansion to the R33 phase. A single priority score will be assigned to each scored application. For the R21/R33 application the initial review group may recommend that only the R21 phase be supported, based on concerns related to the application’s specific goals and the feasibility milestones justifying the expansion to the R33 phase. Deletion of the R33 phase by the review panel or presentation of inadequate milestones in the application may affect the merit rating of the application.
2.A. Additional Review Criteria:

Care and Use of Vertebrate Animals in Research: If vertebrate animals are to
be used in the project, the adequacy of the plans for their care and use will
be assessed. See the Other Research Plan Sections of the PHS398 Research Plan
component of the SF424 (R&R).
Biohazards: If materials or procedures are proposed that are potentially
hazardous to research personnel and/or the environment, determine if the
proposed protection is adequate.
2.B. Additional Review Considerations
Budget
and Period of Support: The reasonableness of the proposed budget and the
appropriateness of the requested period of support in relation to the proposed
research may be assessed by the reviewers. The priority score should not be
affected by the evaluation of the budget.
Applications from Foreign Organizations: Whether the project presents special opportunities for furthering research programs through the use of unusual talent, resources, populations, or environmental conditions in other countries that are not readily available in the United States or that augment existing U.S. resources will be assessed.
2.C. Sharing Research Data
The reasonableness of the data sharing plan or the
rationale for not sharing research data will be assessed by the reviewers.
However, reviewers will not factor the proposed data sharing plan into the
determination of scientific merit or the priority score. The presence of a data
sharing plan will be part of the terms and conditions of the award. The funding
organization will be responsible for monitoring the data sharing policy.

The PHS strongly encourages all grant recipients to
provide a smoke-free workplace and discourage the use of all tobacco products.
In addition, Public Law 103-227, the Pro-Children Act of 1994, prohibits
smoking in certain facilities (or in some cases, any portion of a facility) in
which regular or routine education, library, day care, health care, or early
childhood development services are provided to children. This is consistent
with the PHS mission to protect and advance the physical and mental health of
the American people.
Loan Repayment Programs:
NIH encourages applications for educational loan
repayment from qualified health professionals who have made a commitment to
pursue a research career involving clinical, pediatric, contraception,
infertility, and health disparities related areas. The LRP is an important
component of NIH's efforts to recruit and retain the next generation of
researchers by providing the means for developing a research career unfettered
by the burden of student loan debt. Note that an NIH grant is not required for
eligibility and concurrent career award and LRP applications are encouraged.
The periods of career award and LRP award may overlap providing the LRP
recipient with the required commitment of time and effort, as LRP awardees must
commit at least 50% of their time (at least 20 hours per week based on a 40
