This Funding Opportunity Announcement (FOA) invites cooperative agreement applications that will contribute to a higher resolution understanding of the physical and functional organization of the human islet tissue environment by describing the composition (cellular and molecular) and function of important components of the pancreatic islet and peri-islet tissue architecture, the cell-cell relationships and means of communications used by cell types and cell subtypes within the pancreatic tissue ecosystem, and/or the contribution of adjacent (including acinar, ductal, lymphatic) and neighboring (intestinal, mesenteric and adipose) tissues to islet cell function and dysfunction. Successful projects will integrate the Human Pancreas Analysis Consortium (HPAC), that will consist of the research teams funded in response to this FOA with the Human Pancreas Analysis Program (HPAP), a resource-generation program that was funded in 2016 in response to RFA-DK-15-027. HPAC will become the fifth consortium of the Human Islet Research Network (HIRN, https://hirnetwork.org/ ). HIRN's overall mission is to support innovative and collaborative translational research to understand how human beta cells are lost in T1D, and to find innovative strategies to protect and replace functional beta cell mass in humans. This FOA will only support studies with a primary focus on increasing our understanding of human tissue structure and function, and human disease biology (as opposed to rodent or other animal models). This FOA will not accept applications proposing a clinical trial.

Background
Much remains to be learned about the contribution of the human pancreatic islet tissue environment to Type 1 diabetes pathogenesis. Our inability to easily access the target organ in living individuals, or to extract meaningful biological information from cadaver donor pancreata, has limited our knowledge of pancreatic tissue architecture and function to observations mostly gathered from rodent models. As a result, our understanding of the cellular diversity of the human islet and peri-islet niche, and of the mechanisms by which cell types and cell subtypes communicate with one another in that environment to orchestrate complex endocrine functions, remains limited. Likewise, we know very little of the contribution to islet function and dysfunction of pancreatic compartments that surround the islet (immune, vascular, neuronal, acinar, ductal, lymphatic) or of tissues that are spatially and functionally closely associated with the human pancreas (intestinal, mesenteric, adipose). This lack of knowledge of the basic working components of the human pancreatic tissue impairs our ability to develop therapeutic strategies directed at specific pathogenic or pathological processes, to deliver therapeutic cells or compounds to specific pancreatic sub-compartments, or to develop highly-relevant disease modeling platforms with structural and functional characteristics that closely mimic the endogenous human islet niche. A higher-resolution understanding of the cellular and physical composition of the human pancreatic tissue environment could help us:
The past few years have witnessed the development of powerful technologies that can be applied to the qualitative and quantitative exploration of fixed and live human pancreatic tissues. Single cell analyses of dissociated cells from human pancreata can produce detailed transcriptomics (RNA-seq), epigenomics (ATAC-seq) and multiplexed candidate proteomics (CyTOF) profiles and lead to the identification of cell subtypes that could not be defined previously using combinations of a limited number of known cell surface markers. These complex molecular signatures can then be used to build highly-multiplexed phenotypic assays that, when combined with tissue-scanning/capture protocols and network analysis algorithms, can lead to a spatially resolved characterization of tissues with unprecedented resolution, and enable the interactive, quantitative, and comprehensive exploration of individual cell phenotypes, cell-cell interactions, microenvironments, and morphological structures within intact tissue preparations. In addition to this two-step approach where complex cell signatures are characterized first and later spatially resolved on tissue preparations, some non-targeted analytical methods allow for direct omics measurements in situ using fixed tissue preparations. For example, single-nucleus RNA sequencing from snap frozen pancreata can be combined with 3D spatial reconstruction techniques to explore the cellular diversity of specific pancreatic tissue niches. Other techniques such as mass spectrometry-based MALDI can now be combined with laser capture protocols to provide direct and unbiased proteomics, lipidomics and metabolomics measurements on archived human pancreatic specimens with sub-islet resolution. Finally, progress in the preparation and culture of human pancreatic tissue slices allows for the study of cell-cell interactions and individual cell function in the context of a live 3D tissue environment in which many components of the cytoarchitecture are at least partially preserved (to include neuronal, vascular and ductal compartments). Recent studies using thick pancreatic slices have revealed important differences between the structural and functional architecture of human and mouse pancreata, and suggest a much tighter coupling between the exocrine and endocrine compartments in the human pancreas, including through shared vascular and neuronal networks. Perfused pancreatic tissue slices and other in vitro platforms such as microfluidics-coupled islet bio-mimetics or islets-on-a-chip, can now be used to complement tissue-based and hypothesis-generating omics measurements with mechanistic studies in the context of a live tissue or tissue-like experimental system.
Research Goals and Objectives
This initiative will support the in-depth description of molecular and functional components of the human pancreatic tissue architecture, their establishment during human pancreas development and functional maturation, and their contribution to islet tissue homeostasis and function. This initiative will also support the development and use of innovative phenotypic assays in live human pancreatic cells or tissues (including perfused tissue slices, human islet engraftment models and "islet-on-a-chip" platforms) for mechanistic studies in a live tissue context, and for the manipulation or visualization of regulatory processes that control specific aspects of islet function and dysfunction. Projects in response to this initiative are encouraged to explore the contribution of cell subtypes, tissue microstructures or regulatory pathways to the early stages of human T1D pathogenesis, and to develop molecular tools to manipulate these components of the islet tissue environment as a first step towards the development of novel and highly-targeted regenerative or therapeutic strategies. Proposed approaches could include (but are not limited to):
Given the important differences (genetic, developmental, morphological, cellular and molecular) that exist between the human and rodent islet environments, the use of human pancreatic tissues as a primary experimental system for the acquisition of new knowledge should form the basis of the applications written in response to this initiative. In situations where the efficient exploration of specific aspects of the pancreatic tissue or islet environment is greatly accelerated or facilitated by use of rodent models, limited use of rodent models is allowed if properly justified, but it is expected that results and hypotheses derived from these models will be validated in human tissues in a systematic fashion and throughout the funding period of the grant.
In choosing a source of human tissues, applicants are encouraged to use high-quality repositories, biobanks and procurement networks, whether linked to NIDDK-funded clinical studies such as the Diabetes Prevention Trial-Type 1 (DPT-1) or the Type 1 Diabetes Trialnet (https://www.niddkrepository.org/niddk/home.do , or supported by private research efforts such as the JDRF nPOD (http://www.jdrfnpod.org/ ) or the T1D Exchange (http://www.t1dexchange.org/). If available, pancreata that are either densely genotyped (such as from NIH-supported the GTEx collection: https://commonfund.nih.gov/GTEx) or originating from donors with well-documented disease history or health records, should be used.
The assembly of multidisciplinary teams under a single application to help address the scientific challenges outlined in this initiative is encouraged, particularly to help combine complementary expertise (such as beta cell biology, immunology, experience with state-of-the-art omics technologies or analytical tools, surgical expertise and access to human cadaver donors). Single-investigator applications from individuals with the required expertise to make a major contribution are also welcome.
The HIRN is dedicated to fostering the next generation of diabetes and islet biology investigators; Early Stage Investigators (ESIs) are therefore strongly encouraged to contribute and lead projects in response to this initiative, and to apply as Contact Principal Investigator.
Cooperative Relationships and Data Sharing
Upon funding, successful applicants will integrate with the NIDDK-supported HIRN, whose overall mission is to better understand how human beta cells are lost in T1D, and to find innovative strategies to protect or replace functional beta cell mass in diabetic patients. More specifically, successful applicants will join one of HIRN's five consortia, the Human Pancreas Analysis Consortium (HPAC), that will be composed of the U01 teams funded in response to this FOA and of the Human Pancreas Analysis Program (HPAP) teams, that were funded in 2016 in response to RFA-DK-15-027. HPAP is a resource-generation program that is performing systematic deep phenotyping of the human endocrine pancreas and its interaction with the immune system to better understand the cellular and molecular events that precede and lead to the beta cell loss in type 1 diabetes (T1D). The primary goal of HPAP is to accumulate, analyze, and distribute high value T1D-related datasets to the diabetes research community through an open access database called PANC DB (https://hirnetwork.org/consortium/hpap). Since HPAP investigators are not currently supported to conduct research projects, but specifically to run a systematic and pre-defined series of assays on pancreatic tissues collected from cadaver donors, U01 projects funded through this initiative will constitute the "research component" of the HPAC. All HPAC investigators will be required to work collaboratively with the HPAP teams to find ways of integrating HPAC-generated datasets into PANC DB, and monthly HPAC webinars will be organized to facilitate these collaborative efforts and share recent discoveries.
Regardless of the team-structure proposed in the U01 application, all HPAC investigators will be expected to work closely and collaboratively with their HPAC and HIRN colleagues, and to contribute to an environment of sharing and trust across the network. All methods, reagents, resources, biomaterials, protocols, data and models developed by HPAC investigators are expected to be made available to the research community. Because the individual U01 projects will be coordinated through HPAC, the timeline and processes for sharing within HPAC and with the community at large will be established by the HPAC NIDDK Project Scientist or the HPAC Steering Committee. All participants will be expected to adhere to these policies as a term of the award. Policy documents for HPAC will be accessible on the HIRN website.
As a consortium, HPAC is expected to work closely with the HIRN Administration Hub (HIRN-AH) that is composed of the HIRN Coordinating Center (HIRN-CC) and the Bioinformatics Center (HIRN-BC) to facilitate the organization and sharing of data and reagents generated by HPAC activities. At a minimum, data and analytical tools made available through PANC DB should become easily accessible through the HIRN portal. All HPAC participants will be expected to adhere to the sharing policies developed by the HIRN as a term of the award and to the Cooperative Agreement Terms and Conditions of Award. The NIDDK staff will designate an HPAC investigator to represent HPAC on the HIRN Trans-Network Committee (HIRN-TNC) who will be expected to participate to all meetings of the HIRN-TNC.
HPAC Program Director(s)/Principal Investigator(s) (PD(s)/PI(s)) must participate in person to the annual HIRN Investigator Scientific Meeting, as well as in HPAC Steering Committee teleconferences to be held at least bi-annually. All participants will be obligated to abide by the policies adopted the majority vote of the HPAC Steering Committee. In the application, research project budget requests must include costs for the PD/PI and up to three other members of the individual project to attend the annual HIRN Investigator Scientific Meeting. The annual HIRN Investigator Scientific Meeting will last 2-3 days. In addition to the annual meeting of the HIRN, monthly HPAC webinars will be organized to facilitate early exchange scientific results between HPAC investigators and to update HPAC investigators on HIRN-related activities. All HPAC teleconferences will be organized and administered by the HIRN-CC. Note that the HIRN-CC will support costs of all HPAC and HIRN-related meetings except for costs for research project investigators to travel and attend the meetings. The HIRN-CC is also responsible for providing and maintaining a record of minutes of all HPAC meetings, which will be approved by the HPAC Steering Committee.

NIDDK intends to commit up to $4 million to fund 4-6 awards in FY 2018. The number of awards is contingent upon availability of funds and the submission of a sufficient number of meritorious applications.

Are potential problems, alternative strategies, and benchmarks for success presented? If the project is in the early stages of development, will the strategy establish feasibility and will particularly risky aspects be managed? Have the investigators presented adequate plans to address relevant biological variables, such as sex, for studies in vertebrate animals or human subjects?
Specific to this FOA: Is the project clearly focused on increasing our understanding of human disease biology (as opposed to mouse or other animal models), or at developing knowledge, tools and approaches that could lead to significant translational outcomes in the future? The committee will evaluate the involvement of live vertebrate animals as part of the scientific assessment according to the following criteria: (1) description of proposed procedures involving animals, including species, strains, ages, sex, and total number to be used; (2) justifications for the use of animals versus alternative models and for the appropriateness of the species proposed; (3) interventions to minimize discomfort, distress, pain and injury; and (4) justification for euthanasia method if NOT consistent with the AVMA Guidelines for the Euthanasia of Animals. In accordance with the statutory provisions contained in Section 872 of the Duncan Hunter National Defense Authorization Act of Fiscal Year 2009 (Public Law 110-417), NIH awards will be subject to the Federal Awardee Performance and Integrity Information System (FAPIIS) requirements. Cooperative Agreement Terms and Conditions of Award
The following special terms of award are in addition to, and not in lieu of, otherwise applicable OMB administrative guidelines, HHS grant administration regulations at 45 CFR Parts 74 and 92 (Part 92 is applicable when State and local Governments are eligible to apply), and other HHS PHS, and NIH grant administration policies.
The administrative and funding instrument used for this program will be the cooperative agreement U01, an "assistance" mechanism (rather than an "acquisition" mechanism), in which substantial NIH programmatic involvement with the awardees is anticipated during the performance of the activities. Under the cooperative agreement, the NIH purpose is to support and stimulate the recipients' activities by working jointly with the award recipients in a partnership role; it is not to assume direction, prime responsibility, or a dominant role in the activities. Consistent with this concept, the dominant role and prime responsibility resides with the awardees for the project as a whole, although specific tasks and activities may be shared among the awardees and the NIH as defined below.

Steering Committee
HPAC awardees agree to the governance of the HPAC through a Steering Committee.
HIRN Trans-Network Committee (HIRN-TNC)
The HIRN-TNC will consist of: the PD/PI of the HIRN-CC, the PD/PI of the HIRN-BC, and the Steering Committee Chairs and Project Scientists of the HIRN scientific topic consortia (CHIB, CTAR, CMAI, CBDS and HPAC); the TNC is not a governing body and does not cast votes.
The TNC will facilitate communication and foster collaboration across the different consortia.
The TNC will be responsible for organizing the yearly HIRN Scientific Investigator's Retreat.
The TNC will meet by teleconference at least twice a year and will be organized by the HIRN-CC. Meetings will be used to discuss and prioritize, and review the progress of applications that will use "opportunity pool" funds. Subcommittees of HIRN, as well as working groups for scientific planning may be established and require participation by the HPAC members through in-person, electronic, or teleconference meetings, as appropriate. The HIRN-CC is responsible for providing and maintaining a record of minutes of all EC meetings, which will be approved by the EC.
Expert Scientific Panel (ESP)
An independent panel of 2-5 External Experts will be appointed by the NIDDK and meet by teleconference with the HPAC Project Scientist and the HPAC Project Officer at least once a year. The HPAC-ESP will be updated on progress and give feedback to NIH on adjustments and future directions for the HPAC research projects. On an annual basis, and following input from the ESP members, NIDDK staff will appoint an ESP Chair who will be required to attend the annual HIRN Investigator Scientific Retreat, to participate to the HPAC Steering Committee meetings as ex-officio, and to serve as the HPAC-ESP representative to the larger HIRN-ESP that will also meet once a year. The HPAC ESP Chair will be tasked with relaying the HPAC Steering Committee recommendations for new Opportunity Funds Initiatives to the HIRN-ESP. All HPAC-ESP members will also be invited to listen as ex-officio to HPAC Steering Committee meetings. Members of the HPAC-ESP may be asked, on an ad hoc basis, in the peer review of applications for new research applications that request "opportunity pool" funds. The HIRN-CC will support costs for teleconferences between the ESP and the HPAC Steering Committee, will arrange the HPAC-ESP and HIRN-ESP teleconferences, maintain a record of minutes, and support costs for the HPAC-ESP chair to participate in the annual HIRN Investigator Scientific Retreat.
Dispute Resolution
Disagreements that may arise in scientific/technical matter or programmatic matters (within the scope of the award) between award recipients and the NIDDK may be brought to arbitration after first attempting to resolve the issue through the Steering Committee or its subcommittees, as appropriate. An Arbitration Panel composed of three members will be convened. It will have three members: a designee of the Steering Committee chosen without NIH staff voting, one NIH designee, and a third designee with expertise in the relevant area who is chosen by the other two; in the case of individual disagreement, the first member may be chosen by the individual awardee. This special dispute resolution procedures in no way affect the awardee's right to appeal an adverse action in accordance with PHS regulations at 42 CFR Part 50, Subpart D, and HHS regulations at 45 CFR Part 16.
When multiple years are involved, awardees will be required to submit the Research Performance Progress Report (RPPR) annually and financial statements as required in the NIH Grants Policy Statement.
A final RPPR, invention statement, and the expenditure data portion of the Federal Financial Report are required for closeout of an award, as described in the NIH Grants Policy Statement.
The Federal Funding Accountability and Transparency Act of 2006 (Transparency Act), includes a requirement for awardees of Federal grants to report information about first-tier subawards and executive compensation under Federal assistance awards issued in FY2011 or later. All awardees of applicable NIH grants and cooperative agreements are required to report to the Federal Subaward Reporting System (FSRS) available at www.fsrs.gov on all subawards over $25,000. See the NIH Grants Policy Statement for additional information on this reporting requirement.
In accordance with the regulatory requirements provided at 45 CFR 75.113 and Appendix XII to 45 CFR Part 75, recipients that have currently active Federal grants, cooperative agreements, and procurement contracts from all Federal awarding agencies with a cumulative total value greater than $10,000,000 for any period of time during the period of performance of a Federal award, must report and maintain the currency of information reported in the System for Award Management (SAM) about civil, criminal, and administrative proceedings in connection with the award or performance of a Federal award that reached final disposition within the most recent five-year period. The recipient must also make semiannual disclosures regarding such proceedings. Proceedings information will be made publicly available in the designated integrity and performance system (currently FAPIIS). This is a statutory requirement under section 872 of Public Law 110-417, as amended (41 U.S.C. 2313). As required by section 3010 of Public Law 111-212, all information posted in the designated integrity and performance system on or after April 15, 2011, except past performance reviews required for Federal procurement contracts, will be publicly available.
