Section I. Funding Opportunity Description
Research
Objectives
Purpose
The primary objective of this FOA is to enhance translational research within the mission areas of the NIDCR by harnessing oligonucleotide-based approaches such as RNA interference (RNAi) to modify the expression of genes associated with oral, dental, and craniofacial diseases and disorders. These diseases are complex conditions involving multiple genes and gene-environmental interactions. The ability to selectively silence or modify gene expression is fundamental to understanding complex disease processes and to developing possible therapeutics and prevention strategies. Research proposing to apply RNAi and other oligonucleotide-based strategies can provide rapid and efficient methods for developing new therapeutics for the prevention and treatment of many oral and craniofacial disorders. Applicants should demonstrate that the proposed oligonucleotide-based approaches directly influence molecular, biochemical, electrophysiological or other functional changes in a condition that falls within the NIDCR mission area. These areas include tooth and bone disorders, oral cancer, chronic inflammatory conditions, viral infections, autoimmune disorders, craniofacial birth defects, acute and chronic pain conditions, salivary gland dysfunction, as well as other oral, dental and craniofacial disorders (see http://www.nidcr.nih.gov). Eligible applications may include a component proposing technological innovations to improve the efficiency of delivery, specificity, processing or stability of the oligonucleotide-based strategy, but the proposal must be focused on an oral, dental, or craniofacial health problem. Applications must include a detailed discussion of strategies to assess and minimize nonspecific and off-target effects such as the silencing or induction of unintended targets, as well as potential immune responses triggered by the intervention. Studies that use human samples, animal models, or tissue culture cells are considered appropriate in response to this FOA.
Background
In recent years, the application of oligonucleotide-based methods for modifying gene expression has emerged as a powerful research tool that has vast potential for understanding disease processes and for the development of new therapeutics. These methods have become widely used based on the ease of designing and testing oligonucleotides for any host gene or pathogen whose nucleic acid sequence is known. For example, gene silencing by RNA interference (RNAi) has advanced rapidly in the last decade and several Phase I clinical studies using small interfering RNAs (siRNAs) have been completed successfully. Oligonucleotide-based approaches for altering gene expression include RNAi (siRNA, shRNA, and miRNA), splice and translation blocking morpholinos, aptamers, as well as more recent techniques such as chromosome-targeted small duplex RNAs that can activate as well as repress gene expression. The possible therapeutic applications of oligonucleotide-based methods are broad and far-reaching, ranging from acquired diseases, such as viral infections, to genetic disorders.
The main obstacles to applying RNAi as a therapeutic approach have been the problems of achieving effective intracellular in vivo delivery into mammalian cells, and achieving efficient and stable target gene inhibition. Considerable progress has been made recently in overcoming these problems and strategies such as cholesterol-conjugation have been shown to greatly facilitate siRNA uptake. The mucosal surfaces of the body have proven to be one of the most accessible routes for siRNA delivery. Efficient uptake of siRNA has been demonstrated in the respiratory tract after intranasal topical application and in the cervix and vagina after topical application of siRNA-lipid complexes. These findings suggest that the oral mucosa may also present an accessible portal of entry after topical application or direct injection of siRNAs. Research is needed to assess if oligonucleotide-based methods can be used for the treatment of inflammatory oral diseases, mucosal malignancies such as head and neck cancers and other oral health conditions. Studies are also required to identify which primary cell types in mucosal tissues mediate uptake of siRNA and which cells are amenable to therapeutic interventions. Finally, research is needed for the optimization of specificity and stability of RNAi-mediated functional effects in oral tissues. For direct therapeutic modalities, consideration should be given to possible adverse events arising from off-target effects (general toxicity) as well as dosing and pharmacokinetic parameters.
Examples of research topics and experimental approaches that are appropriate for this FOA include, but are not limited to:
The R21 mechanism is intended to encourage new exploratory and developmental research projects. For example, such projects could assess the feasibility of a novel area of investigation or a new experimental system that has the potential to enhance health-related research. Another example could include the unique and innovative use of an existing methodology to explore a new scientific area. These studies may involve considerable risk but may lead to a breakthrough in a particular area, or to the development of novel techniques, agents, methodologies, models, or applications that could have a major impact on a field of biomedical, behavioral, or clinical research.
Applications for R21 awards should describe projects distinct from those supported through the traditional R01 mechanism. For example, long-term projects, or projects designed to increase knowledge in a well-established area, will not be considered for R21 awards. Applications submitted under this mechanism should be exploratory and novel. These studies should break new ground or extend previous discoveries toward new directions or applications.

II. Award Information
1. Mechanism of Support
This Funding Opportunity Announcement (FOA) will use the NIH
Exploratory/Developmental Grant (R21) award mechanism. The
applicant(s) will be solely responsible for planning, directing, and executing
the proposed project.
This FOA uses Just-in-Time information concepts. It also uses the modular budget formats (see the Modular Applications and Awards section of the NIH Grants Policy Statement. Specifically, if you are submitting an application with direct costs in each year of $250,000 or less (excluding consortium Facilities and Administrative [F&A] costs), use the PHS398 Modular Budget component provided in the SF424 (R&R) Application Package and SF424 (R&R) Application Guide (see specifically Section 5.4, Modular Budget Component, of the Application Guide).

The R21 mechanism is for new projects only. Competing renewals (formerly competing continuation) will not be accepted.
2. Funds Available
Because the nature and scope of the proposed research will vary from
application to application, it is anticipated that the size and duration of
each award will also vary. Although the financial plans of the Institutes and
Centers (ICs) provide support for this program, awards pursuant to this funding
opportunity are contingent upon the availability of funds and the submission of
a sufficient number of meritorious applications.
The total project period for an application submitted in response to this funding opportunity may not exceed 2 years. Although the size of award may vary with the scope of research proposed, it is expected that applications will stay within the budgetary guidelines for an exploratory/developmental project. Direct costs are limited to $275,000 over a two-year period, with no more than $200,000 in direct costs allowed in any single year. Applicants may request direct costs in $25,000 modules, up to the total direct costs limitation of $275,000 for the combined two-year award period.
The participating organization NIDCR intends to commit a total of approximately $2,500,000 dollars over two years, in FY2008 and FY2009, to fund approximately 5-10 applications.
NIH grants policies as described in the NIH Grants Policy Statement will apply to the applications submitted and awards made in response to this FOA.
Facilities
and Administrative (F&A) costs requested by consortium participants are not

Any individual with the skills, knowledge, and resources necessary to carry out the proposed research as the Project Director/Principal Investigator (PD/PI) is invited to work with his/her organization to develop an application for support. Individuals from underrepresented racial and ethnic groups as well as individuals with disabilities are always encouraged to apply for NIH support.
More than one PD/PI, or multiple PDs/PIs, may be designated on the application for projects that require a team science approach that clearly does not fit the single-PD/PI model. SPECIAL INSTRUCTIONS
Applications with Multiple PDs/PIs
When multiple PDs/PIs are proposed, NIH requires one PD/PI to be designated as the "Contact PI, who will be responsible for all communication between the PDs/PIs and the NIH, for assembling the application materials outlined below, and for coordinating progress reports for the project. The contact PD/PI must meet all eligibility requirements for PD/PI status in the same way as other PDs/PIs, but has no other special roles or responsibilities within the project team beyond those mentioned above.
Information for the Contact PD/PI should be entered in item 15 of the SF424(R&R) Cover component. All other PDs/PIs should be listed in the Research & Related Senior/Key Person component and assigned the project role of PD/PI. Please remember that all PDs/PIs must be registered in the eRA Commons prior to application submission. The Commons ID of each PD/PI must be included in the Credential field of the Research & Related Senior/Key Person component. Failure to include this data field will cause the application to be rejected.
All projects proposing Multiple PDs/PIs will be required to include a new section describing the leadership of the project.
Multiple PD/PI Leadership Plan: For applications designating multiple PDs/PIs, a new section of the research plan, entitled Multiple PD/PI Leadership Plan [Section 14 of the Research Plan Component in the SF424 (R&R)], must be included. A rationale for choosing a multiple PD/PI approach should be described. The governance and organizational structure of the leadership team and the research project should be described, including communication plans, process for making decisions on scientific direction, and procedures for resolving conflicts. The roles and administrative, technical, and scientific responsibilities for the project or program should be delineated for the PDs/PIs and other collaborators.
If budget allocation is planned, the distribution of resources to specific components of the project or the individual PDs/PIs should be delineated in the Leadership Plan. In the event of an award, the requested allocations may be reflected in a footnote on the Notice of Award.
Applications Involving a Single Institution

Applications Involving Multiple Institutions
When multiple institutions are involved, one institution must be designated as the prime institution and funding for the other institution(s) must be requested via a subcontract to be administered by the prime institution. When submitting a detailed budget, the prime institution should submit its budget using the Research & Related Budget component. All other institutions should have their individual budgets attached separately to the Research & Related Subaward Budget Attachment(s) Form. Funding Restrictions
All NIH awards are subject to the terms and
conditions, cost principles, and other considerations described in the NIH Grants Policy Statement.
Pre-award costs are allowable. A grantee may, at
its own risk and without NIH prior approval, incur obligations and expenditures
to cover costs up to 90 days before the beginning date of the initial budget
period of a new award if such costs: are necessary to conduct the project, and
would be allowable under the grant, if awarded, without NIH prior approval. If
specific expenditures would otherwise require prior approval, the grantee must
obtain NIH approval before incurring the cost. NIH prior approval is required
for any costs to be incurred more than 90 days before the beginning date of the
initial budget period of a new award.
The incurrence of pre-award costs in anticipation of a
competing or non-competing award imposes no obligation on NIH either to make
the award or to increase the amount of the approved budget if an award is made
for less than the amount anticipated and is inadequate to cover the pre-award
costs incurred. NIH expects the grantee to be fully aware that pre-award costs
result in borrowing against future support and that such borrowing must not
impair the grantee's ability to accomplish the project objectives in the
approved time frame or in any way adversely affect the conduct of the project.

The NIH R21 exploratory/developmental grant is a mechanism for supporting novel scientific ideas or new model systems, tools, or technologies that have the potential to significantly advance our knowledge or the status of health-related research. Because the Research Plan is limited to 15 pages, an exploratory/developmental grant application need not have extensive background material or preliminary information as one might normally expect in an R01 application. Accordingly, reviewers will focus their evaluation on the conceptual framework, the level of innovation, and the potential to significantly advance our knowledge or understanding. Reviewers will place less emphasis on methodological details and certain indicators traditionally used in evaluating the scientific merit of R01 applications, including supportive preliminary data. Appropriate justification for the proposed work can be provided through literature citations, data from other sources, or, when available, from investigator-generated data. Preliminary data are not required for R21 applications in response to this FOA; however, they may be included if available.
The goals of NIH supported research are to advance our understanding of biological systems, to improve the control of disease, and to enhance health. In their written critiques, reviewers will be asked to comment on each of the following criteria in order to judge the likelihood that the proposed research will have a substantial impact on the pursuit of these goals. Each of these criteria will be addressed and considered in assigning the overall score, weighting them as appropriate for each application.
Note that an application does not need to be strong in all
categories to be judged likely to have major scientific impact and thus deserve
a high priority score. For example, an investigator may propose to carry out
important work that by its nature is not innovative but is essential to move a
field forward.
Significance: Does this study address an
important problem? If the aims of the application are achieved, how will
scientific knowledge or clinical practice be advanced? What will be the effect
of these studies on the concepts, methods, technologies, treatments, services,
or preventative interventions that drive this field?
Approach: Are the conceptual or clinical
framework, design, methods, and analyses adequately developed, well integrated,
well reasoned, and appropriate to the aims of the project? Does the applicant
acknowledge potential problem areas and consider alternative tactics? For applications designating multiple PDs/PIs, is
the leadership approach, including the designated roles and responsibilities,
governance, and organizational structure, consistent with and justified by the
aims of the project and the expertise of each of the PDs/PIs?
Innovation: Is the project
original and innovative? For example: Does the project challenge existing
paradigms or clinical practice; address an innovative hypothesis or critical
barrier to progress in the field? Does the project develop or employ novel
concepts, approaches, methodologies, tools, or technologies for this area?
Investigators: Are the PD/PI(s)
and other key personnel appropriately trained
and well suited to carry out this work? Is the work proposed appropriate to the
experience level of the principal investigator and other researchers? Does the
PD/PI(s) and investigative team bring complementary and integrated expertise to
the project (if applicable)?
Environment: Do(es) the scientific environment(s) in which the work will
be done contribute to the probability of success? Do the proposed studies
benefit from unique features of the scientific environment, or subject
populations, or employ useful collaborative arrangements? Is there evidence of
institutional support?

Resubmission
Applications (formerly revised/amended applications): Are the responses
to comments from the previous scientific review group adequate? Are the

Protection of Human Subjects from Research Risk: The involvement of human subjects and protections from research risk relating
to their participation in the proposed research will be assessed. See the

Inclusion of Women, Minorities and Children in Research: The adequacy of
plans to include subjects from both genders, all racial and ethnic groups (and
subgroups), and children as appropriate for the scientific goals of the
research will be assessed. Plans for the recruitment and retention of subjects

Care and Use of Vertebrate Animals in Research: If vertebrate animals
are to be used in the project, the adequacy of the plans for their care and use

Biohazards: If materials or procedures are proposed that are potentially
hazardous to research personnel and/or the environment, determine if the
proposed protection is adequate.
2.B. Additional Review Considerations
Budget and Period of Support: The reasonableness of the proposed budget and the
appropriateness of the requested period of support in relation to the proposed
research may be assessed by the reviewers. The priority score should not be
affected by the evaluation of the budget.
Applications from Foreign Organizations: Whether the project presents special opportunities for furthering research programs through the use of unusual talent, resources, populations, or environmental conditions in other countries that are not readily available in the United States or that augment existing U.S. resources will be assessed.
