Funding Opportunity Purpose |
The purpose of this funding opportunity is to stimulate the adoption and validation of novel powerful single cell analysis (SCA) approaches by supporting collaborations of currently funded U01 investigators with developers of the SCA approaches. The collaborations must be new, possess the potential to substantially further the aims of the U01 project, and provide iterative and informed feedback to the SCA developers for continued refinement. By strengthening the interface between users and developers, the objective is to accelerate the discovery of fundamental new biological insights and translation of new diagnostic and therapeutic methods that analysis at the single cell level will allow. The purpose of this funding opportunity is to stimulate the adoption and scientific validation of novel single cell approaches by supporting new collaborative efforts. The collaborative efforts must advance the aims of a funded U01 project through the application of single cell approaches still in a developmental stage and provide opportunities for iterative feedback to the collaborators developing the single cell analysis approaches. Revision applications (formerly referred to as competitive supplements) are sought from U01 grantees/investigators who propose to add and test a new component involving single cell analysis to their research project. Collaborations of U01 investigators with investigators currently funded through the Common Fund Single Cell Analysis Program (http://commonfund.nih.gov/singlecell/fundedresearch.aspx) are strongly encouraged. The primary goal then is to strengthen the interactions between single cell methods developers and end users and thereby accelerate the pace at which these emerging methods become broadly useful to the biomedical community.
BackgroundThis initiative is part of the Single Cell Analysis Program (SCAP) and is funded through the NIH Common Fund (See http://nihroadmap.nih.gov/), which supports cross-cutting programs that are expected to have exceptionally high impact. Common Fund initiatives address key roadblocks in biomedical research that impede basic scientific discovery and its translation into improved human health. In addition, these programs capitalize on emerging opportunities to catalyze progress across multiple biomedical fields.
Single cell analysis is emerging as an important field of research as technologies improve in sensitivity and with increased throughput to allow measurement and understanding of cell heterogeneity in complex biological systems. It is becoming clearer that behavior at the single cell level can have profound effects at higher levels of organization. By profiling individual cells it is possible to resolve rare cells, transient cell states, and emergent properties, as well as the influence of organization and environment on such cells and states, which cannot be described by ensemble measurements. The long-term goal of the SCAP is to accelerate this move towards personalizing health to the cellular level by understanding the link between cell heterogeneity, tissue function and emergence of disease through the discovery, development and translation of innovative approaches which will dramatically change the way cells are characterized.
The SCAP focuses on supporting work which will systematically measure, analyze and model cell-to-cell variation, and identify crucial differences and rare biological states, which may have important functional consequences. To robustly and systematically describe cell level heterogeneity, projects are encouraged to take a multiplexed approach with minimal perturbation where possible. Technologies and methods that capture spatiotemporal information to understand how individual cells function as part of a population are of interest. In addition, the SCAP emphasizes the application of new technologies to in situ populations of cells from multicellular organisms to link cell state measurements with complex, functional tissues which can inform our understanding of disease processes.

Objectives
The goal of the Single Cell Analysis Program is to accelerate the discovery, development and translation of cross-cutting, innovative tools and approaches to understand functional cell heterogeneity in biologically relevant cell populations using single cell analytical techniques. This announcement contributes to this goal by providing an opportunity for scientists to extend their own funded research projects by applying innovative single cell approaches and for single cell analysis methods developers to field test and refine their methods. The supplemental funds should be used for demonstration projects to 1) test the utility of new single cell approaches; 2) advance the aims of the parent grant; and 3) forge collaborations among scientists with complementary expertise. Responsive applications will address all three points. The parent U01 project should not have a single cell component in the current funded research plan and the Program Director/Principal Investigator (PD/PI) should have no (or only limited) expertise in single cell approaches. Consistent with the broader goals of the SCAP, the intent of this funding opportunity is to support the analysis of single eukaryotic cells in biologically relevant contexts including in vivo, explant or in situ preparations, or other contexts where complex tissue (or tissue-like) architecture is present. Single cell experiments using dispersed cell cultures will not be permitted with the exception of human induced pluripotent stem cell (iPSC) lines or human embryonic stem cell (hESC) lines that are approved for NIH funding through the NIH registry (http://stemcells.nih.gov/research/registry/Pages/Default.aspx).
The efforts encouraged through this FOA are project specific. The NIH would especially like to foster and encourage new collaborations among scientists, trainees, engineers and computational biologists with complementary expertise, who may not have worked together previously. For example, applicants may partner with single cell technology developers, bioengineers, or others with experience using and applying single cell tools, including computational approaches. Key personnel on funded revisions and/or the parent grant are expected to attend the annual Common Fund Single Cell Analysis workshop meeting held in the Bethesda area.
Funded U01 projects with existing single cell analysis components are excluded. Since the goal is to test and promote novel single cell technologies and approaches in areas that have not benefited from these tools previously, applications where the PD/PI of the parent grant has a publication record of using single cell approaches in recent years may be considered non-responsive.

Protections for Human Subjects
For research that involves human subjects but does
not involve one of the six categories of research that are exempt under 45 CFR
Part 46, the committee will evaluate the justification for involvement of human
subjects and the proposed protections from research risk relating to their
participation according to the following five review criteria: 1) risk to
subjects, 2) adequacy of protection against risks, 3) potential benefits to the
subjects and others, 4) importance of the knowledge to be gained, and 5) data
and safety monitoring for clinical trials.
For research that involves human subjects and meets the criteria for one or
more of the six categories of research that are exempt under 45 CFR Part 46,
the committee will evaluate: 1) the justification for the exemption, 2) human
subjects involvement and characteristics, and 3) sources of materials. For

Inclusion of Women, Minorities, and Children

Vertebrate Animals
The committee will evaluate the involvement of live vertebrate animals as part of the scientific assessment according to the following five points: 1) proposed use of the animals, and species, strains, ages, sex, and numbers to be used; 2) justifications for the use of animals and for the appropriateness of the species and numbers proposed; 3) adequacy of veterinary care; 4) procedures for limiting discomfort, distress, pain and injury to that which is unavoidable in the conduct of scientifically sound research including the use of analgesic, anesthetic, and tranquilizing drugs and/or comfortable restraining devices; and 5) methods of euthanasia and reason for selection if not consistent with the AVMA Guidelines on Euthanasia. For Revisions, the committee will consider the appropriateness of the proposed expansion of the scope of the project. If the Revision application relates to a specific line of investigation presented in the original application that was not recommended for approval by the committee, then the committee will consider whether the responses to comments from the previous scientific review group are adequate and whether substantial changes are clearly evident. Has sufficient progress been made in the parent project to justify the proposed expansion of scope?

Applications from Foreign Organizations
Reviewers will assess whether the project presents special opportunities for furthering research programs through the use of unusual talent, resources, populations, or environmental conditions that exist in other countries and either are not readily available in the United States or augment existing U.S. resources.
Select Agent Research
Reviewers will assess the information provided in this section of the application, including 1) the Select Agent(s) to be used in the proposed research, 2) the registration status of all entities where Select Agent(s) will be used, 3) the procedures that will be used to monitor possession use and transfer of Select Agent(s), and 4) plans for appropriate biosafety, biocontainment, and security of the Select Agent(s).

Cooperative Agreement Terms and Conditions of Award
NIH Cooperative Agreements are subject to terms and conditions in addition to, and not in lieu of, otherwise applicable U.S. Office of management and Budgets (OMB) Administrative Guidelines, U.S. Department of Health and Human Services (DHHS) grant administration regulations at 45 CFR Parts 74 and 92 (Part 92 is applicable when State and local governments are eligible to apply), and other HHS, PHS, and NIH grants administration policies.
Each revision made under this FOA will be subject to the same terms and conditions specified for the original U01 award. Applicants should consult the Notice of Award for their current U01 for the specifics of those terms and conditions.
When multiple years are involved, awardees will be required to submit the annual Non-Competing Progress Report (PHS 2590 or RPPR) and financial statements as required in the NIH Grants Policy Statement.
A final progress report, invention statement, and the expenditure data portion of the Federal Financial Report are required for closeout of an award, as described in the NIH Grants Policy Statement.
The Federal Funding Accountability and Transparency Act of 2006 (Transparency Act), includes a requirement for awardees of Federal grants to report information about first-tier subawards and executive compensation under Federal assistance awards issued in FY2011 or later. All awardees of applicable NIH grants and cooperative agreements are required to report to the Federal Subaward Reporting System (FSRS) available at www.fsrs.gov on all subawards over $25,000. See the NIH Grants Policy Statement for additional information on this reporting requirement.
