FOA Purpose |
This Funding Opportunity Announcement (FOA) issued by the National Institutes of Health, encourages applications that will develop novel technologies and/or tools to facilitate the study of genes and proteins at the synapse on a large scale. Being able to characterize the synaptome in the context of hundreds of genes/proteins would be a major advance for basic and translational research. The technologies/tools created would be expected to affect the fields of neurology, psychiatry, and neuroscience by enriching existing resources as well as those in development. The new tools should enable discovery science related to the synapse by providing for substantially greater sensitivity, selectivity, spatiotemporal resolution and markedly decreased costs compared to currently available technologies. |

The NIH Blueprint for Neuroscience Research is a framework to enhance cooperative activities among 16 NIH Institutes, Centers and Offices that jointly support neuroscience-related research, with the aim of accelerating discoveries and reducing the burden of nervous system disorders (for further information, see http://neuroscienceblueprint.nih .gov/). This Funding Opportunity Announcement (FOA) encourages applications directed toward the development of novel and/or vastly improved means for global analysis of the synapse. Given the unique compartmentalized nature of the nervous system, "omics" approaches to analyze this system have inherent additional difficulties such as ability to isolate specific cell types or even subcellular entities of interest such as the synapse. Tools and technologies for the study of subcellular or cell type specific molecular/genetic entities contributing to neurotransmission and neural plasticity would offer compelling new information related to neural function and dysfunction. Studies in invertebrates and vertebrates are welcome if the resulting resources will accelerate research in the area of synapse biology.
Over the previous decade, technological developments in the life sciences have led to a proliferation of "omics" approaches, whose goals involve comprehensive discovery of entire classes of biochemical or genetic entities for a given tissue or cell type. The nervous system represents a challenge and also an opportunity for this type of approach because of its unique cellular and subcellular compartmentalization. A variety of methods are now available (or in development) for isolating components from different neural cell types and subcellular domains. The goal of this FOA is to harness and/or develop such methodologies for the purpose of comprehensive identification of genes, proteins or other biochemical analytes directly contributing to synaptic structure or function.
Much evidence exists that points to specific pathologies related to the synapse in a number of brain disorders, including autism, schizophrenia and other neurodevelopmental and neurodegenerative disorders. The complexity of the synapse and its molecular diversity is vast. Through this FOA, the NIH seeks applications that propose to develop novel tools that will further our understanding of synaptic function as it relates to plasticity, cell type specificity, neurodevelopment and dysfunction of brain circuitry. Tools to interrogate the synapse could include chip-based approaches to characterize gene or protein expression, protein capture, or novel technologies for biochemical or genetic analysis. These technologies should provide resources relevant to many of the existing Blueprint programs, especially neuroplasticity, cognition, disease research and therapeutic target discovery.
A cell's identity can be characterized in a number of ways including its lineage, molecular composition, epigenetic state, activity profile, morphology and the nature of its connections (inputs and outputs). Specifically, the ratio of excitatory vs. inhibitory inputs, the specific location of synapses as well as the strength and capacity for plasticity can provide useful information about potential function. Furthermore, the dynamical nature of synapses must be considered when attempting to understand their role in circuit function.
Recent advances in genetic engineering technology are enabling investigators to specifically label discrete synapses in the mammalian brain, allowing a fine-scale study of synaptic biology. These approaches use the Cre/LoxP system (or a variant thereof) to target synaptic protein markers and to provide visualization through fluorescently labeled proteins. Spatially targeting the synapse using genetic engineering coupled with omics based approaches to study the genome, epigenome, transcriptome and proteome would provide a wealth of novel information on synaptic biology.
This FOA seeks applications to develop novel tools and/or technology that will enable broader assessment of the synaptome. Improvements to existing tools that substantially increase efficiency and accuracy, greatly improve detection and/or resolution, substantially reduce costs or increase throughput are also encouraged.
Examples of research topics include, but are not limited to, those developing or significantly improving:
For protein assessments, a number of measures of the biochemical state of groups of proteins might be proposed, (including myristoylation and palmitoylation, protein acetylation, methylation, crotonylation, malonylation, phosphorylation, glycosylation, sialylation, etc.). In addition, measurements of the various facets of protein interactive states with other existing synaptic or cytoskeletal proteins might be proposed.
Research designs should focus on tool development, but tools may be tested in the context of biological systems. Examples of projects using biological systems include: the study of differences between two synapse types from the same organism; and comparing spatiotemporal differences in synapses that have changed due to learning or environmental perturbations. Applicants are expected to explain the significance of the specific technical approach with respect to current state of the art techniques. However, any proposed biological assay(s) should be chosen strictly for utility (rather than biological novelty) in proof-of-concept testing of the innovative technique.
Applications that propose primarily hypothesis-driven studies to answer specific biology questions, rather than to develop or improve a tool or technology to investigate synapses will be considered non-responsive to the FOA.
In order to maximize the impact of these advances, applicants are strongly encouraged to include a Dissemination Plan describing how they will share research resources, data, and model organisms developed in response to this announcement. The NIH expects that any model organisms developed with support from this FOA will be submitted to an appropriate central repository, or that a compelling explanation as to why this is not possible will be included in the model organism sharing plan.
NIH understands and accepts that early stage, high-risk/high-impact applications that are responsive to this solicitation will be much riskier than for most other solicitations.

Only the review criteria described below will be considered in the review process. As part of the NIH mission, all applications submitted to the NIH in support of biomedical and behavioral research are evaluated for scientific and technical merit through the NIH peer review system.
The R21 exploratory/developmental grant supports investigation of novel scientific ideas or new model systems, tools, or technologies that have the potential for significant impact on biomedical or biobehavioral research. An R21 grant application need not have extensive background material or preliminary information. Accordingly, reviewers will focus their evaluation on the conceptual framework, the level of innovation, and the potential to significantly advance our knowledge or understanding. Appropriate justification for the proposed work can be provided through literature citations, data from other sources, or, when available, from investigator-generated data. Preliminary data are not required for R21 applications; however, they may be included if available.
Reviewers will provide an overall impact/priority score to reflect their assessment of the likelihood for the project to exert a sustained, powerful influence on the research field(s) involved, in consideration of the following review criteria and additional review criteria (as applicable for the project proposed).
Reviewers will consider each of the review criteria below in the determination of scientific merit, and give a separate score for each. An application does not need to be strong in all categories to be judged likely to have major scientific impact. For example, a project that by its nature is not innovative may be essential to advance a field.
Significance
Does the project address an important problem or a critical barrier to progress in the field? If the aims of the project are achieved, how will scientific knowledge, technical capability, and/or clinical practice be improved? How will successful completion of the aims change the concepts, methods, technologies, treatments, services, or preventative interventions that drive this field? If fully successful, will the proposed work significantly contribute to tools for analyzing synaptic function and synaptic transmission in health and disease? Is the new technology likely to be widely-adopted by the neuroscience research community or is it likely to be adopted by only a small number of users?
Investigator(s)
Are the PD(s)/PI(s), collaborators, and other researchers well suited to the project? If Early Stage Investigators or New Investigators, or in the early stages of independent careers, do they have appropriate experience and training? If established, have they demonstrated an ongoing record of accomplishments that have advanced their field(s)? If the project is collaborative or multi-PD(s)/PI(s), do the investigators have complementary and integrated expertise; are their leadership approach, governance and organizational structure appropriate for the project?
Innovation
Does the application challenge and seek to shift current research or clinical practice paradigms by utilizing novel theoretical concepts, approaches or methodologies, instrumentation, or interventions? Are the concepts, approaches or methodologies, instrumentation, or interventions novel to one field of research or novel in a broad sense? Is a refinement, improvement, or new application of theoretical concepts, approaches or methodologies, instrumentation, or interventions proposed? How innovative is the proposed work? If fully successful, does the new technology have the potential to transform our analysis of synapses or will it lead to merely incremental improvements?
Approach
Are the overall strategy, methodology, and analyses
well-reasoned and appropriate to accomplish the specific aims of the project?
Are potential problems, alternative strategies, and benchmarks for success
presented? If the project is in the early stages of development, will the
strategy establish feasibility and will particularly risky aspects be
managed?
If the project involves clinical research, are the plans for 1) protection of
human subjects from research risks, and 2) inclusion of minorities and members
of both sexes/genders, as well as the inclusion of children, justified in terms
of the scientific goals and research strategy proposed?
Environment
Will the scientific environment in which the work will be done contribute to the probability of success? Are the institutional support, equipment and other physical resources available to the investigators adequate for the project proposed? Will the project benefit from unique features of the scientific environment, subject populations, or collaborative arrangements?
As applicable for the project proposed, reviewers will evaluate the following additional items while determining scientific and technical merit, and in providing an overall impact/priority score, but will not give separate scores for these items.
Protections for Human Subjects
For research that involves human subjects but does
not involve one of the six categories of research that are exempt under 45 CFR
Part 46, the committee will evaluate the justification for involvement of human
subjects and the proposed protections from research risk relating to their
participation according to the following five review criteria: 1) risk to
subjects, 2) adequacy of protection against risks, 3) potential benefits to the
subjects and others, 4) importance of the knowledge to be gained, and 5) data
and safety monitoring for clinical trials.
For research that involves human subjects and meets the criteria for one or

Vertebrate Animals
The committee will evaluate the involvement of live vertebrate animals as part of the scientific assessment according to the following five points: 1) proposed use of the animals, and species, strains, ages, sex, and numbers to be used; 2) justifications for the use of animals and for the appropriateness of the species and numbers proposed; 3) adequacy of veterinary care; 4) procedures for limiting discomfort, distress, pain and injury to that which is unavoidable in the conduct of scientifically sound research including the use of analgesic, anesthetic, and tranquilizing drugs and/or comfortable restraining devices; and 5) methods of euthanasia and reason for selection if not consistent with the AVMA Guidelines on Euthanasia.
