Section I. Funding Opportunity Description
1. Research Objectives
Purpose
The purpose of this Funding Opportunity Announcement (FOA) is to stimulate the use of state-of-the-art electrophysiological, molecular, genetic and/or imaging techniques in animals to examine the neural mechanisms by which regions in the mature and developing prefrontal cortex (PFC) interact with other cortical and sub cortical systems to give rise to complex mental functions (e.g., cognition, emotion, reward, motivation). This FOA seeks applications employing sophisticated approaches that enable precise spatio-temporal manipulation of circuits to better understand how the prefrontal cortex causally influences the function and development of these brain networks. It is expected that applications submitted in response to this FOA will use non-human primates where the anatomical and functional homology of the prefrontal cortex with humans has been relatively well established. The use of rodents to develop novel approaches to manipulate prefrontal circuitry and examine associated complex behaviors will be considered if these techniques have the clear potential for near-term translation to non-human primates. Identifying how the PFC regulates the circuits that support complex cognitive and emotional functions is critically important for discovering the pathophysiology of mental disorders and for future treatment development.
Background
Most psychiatric disorders are characterized by significant impairments in cognitive and emotional control, functions attributed in part to the prefrontal cortex (PFC). These deficits often persist even with treatment and significantly impact the ability to function in everyday life situations. Most cognitive and emotional impairments can be correlated with abnormalities in the anatomy and/or neural activity within regions in and around the PFC including the lateral and dorsolateral PFC, orbital frontal cortex, and the anterior cingulate region. Over the past several decades, single-unit electrophysiological, functional imaging, and lesion studies have provided a great deal of information about the connectivity and properties of these regions. Recent findings from a range of studies suggest that regions within the prefrontal cortex can exert a significant and surprisingly strong influence on the generation or cessation of behaviors relevant to mental health and disease. For example, fear extinction studies in rats point to infralimbic (prefrontal cortex) modulation of the amygdala; studies on learned helplessness and resilience point to the role of the same infralimbic regions in modulating the function of the dorsal raphe nucleus. Recent work in humans suggests that deep brain electrical stimulation of medial prefrontal cortex (subgenual anterior cingulate cortex) can result in relief from severe depression in some patients. Finally, research using human and non-human primates has uncovered a network of neocortical regions, including the prefrontal cortex, that is involved in decision-making. The results of these studies suggest that interactions between circuits in the PFC and other cortical and subcortical systems are critical for normal cognitive and emotional function. However, much less is known about the anatomical and neural mechanisms by which prefrontal regions exert these functional influences.
Understanding how regions within the prefrontal cortex causally influence the function of other brain regions and circuits requires a detailed understanding of the anatomical targets of prefrontal cortical neurons and the ability to manipulate interconnected circuits to understand how their output influences the physiological properties of neurons within other regions in the circuit. Similarly, there is a critical need to identify the involvement of the prefrontal cortex and related circuits in the emergence of particular behavioral and cognitive functions during development. Identifying these circuits and understanding how they functionally interact in both developing and adult animals to support complex cognitive and emotional functions is a critical next step for the discovery of the pathophysiology of mental disorders and will enable the development of more effective and targeted interventions for psychiatric disorders.
Studies to be supported
The goal of this initiative is to move beyond studies of individual brain regions and to adopt a systems-wide approach to understanding the development and function of the prefrontal cortex in the context of other brain regions that regulate cognition, emotion, and other complex behaviors (e.g., hippocampus, striatum, and limbic regions). Innovative technologies to be supported under this FOA include multi-site, high density simultaneous electrophysiological recordings, imaging in awake animals, genetically or pharmacologically enabled cell and/or circuit activation and inactivation, and the use of molecular tracers and activity sensors (e.g., viral vectors and florescent protein tags) to map (spatially and developmentally) specific circuits and sub circuits that support PFC-mediated behaviors. Applications submitted in response to this FOA are expected to use a combination of these techniques that allows for the precise manipulation of specific circuits and an analysis of the effects on the function of connected brain regions and on behavior. Collaborations between molecular biologists developing genetic tools in rodents and systems neuroscientists examining the circuits that support complex behavior in non-human primates are strongly encouraged. It is expected that applications funded under this initiative will use non-human primates where the anatomical and functional homology of the PFC with humans has been relatively well established and invasive studies of complex cognitive function are possible. The use of rodents to develop novel approaches to manipulate prefrontal circuitry and examine associated complex behaviors will be considered if these techniques have the clear potential for near-term translation to non-human primates. Given the complexity of the data likely to be generated, it is expected that all applications will include the use of sophisticated analytical techniques to understand the dynamic interactions between different regions as well as the relationship between neural activity and behavior.
The following approaches and topics would be appropriate for this FOA. This list is not meant to be exhaustive but can provide guidance on the types of projects that might be supported under this FOA.
For all approaches, it will be important to define the experimental question and identify the limitations and advantages of the available methods. Because this FOA emphasizes the use of novel approaches and combinations of state-of-the-art techniques, it is not expected that applications will contain substantial preliminary data. However, applicants are encouraged to include any data demonstrating feasibility of their research approach.
In summary, there have been recent technical advances that have resulted in the development of powerful new approaches for manipulating receptors, cells, and circuits at increasingly high spatial and temporal resolutions. Multi-site recording techniques combined with electrical and pharmacological inactivation and stimulation are being used effectively in both rodents and primates. Genetic and molecular approaches for manipulating circuits have been successfully used in rodents and these techniques are now being applied to non-human primates. Accordingly, the field is now poised to adapt the use of these tools in non-human primates that are capable of complex behavior. Significant advances in understanding prefrontal influences on the circuits that support complex mental function will require collaborations between teams of investigators that can merge novel technical approaches with studies of cognitive and emotional behavior. Identifying how the PFC regulates the circuits that support complex cognitive and emotional functions is critically important for discovering the pathophysiology of mental disorders. Furthermore, understanding the neural circuits that causally mediate cognition and emotion will enable the development of more effective and targeted interventions for psychiatric disorders.
The evolution and vitality of the biomedical sciences require a constant infusion of new ideas, techniques, and points of view. These may differ substantially from current thinking or practice and may not yet be supported by substantial preliminary data. By using the R21 mechanism, the NIH seeks to foster the introduction of novel scientific ideas, model systems, tools, agents, targets, and technologies that have the potential to substantially advance biomedical research.
The R21 mechanism is intended to encourage new exploratory and developmental research projects. For example, such projects could assess the feasibility of a novel area of investigation or a new experimental system that has the potential to enhance health-related research. Another example could include the unique and innovative use of an existing methodology to explore a new scientific area. These studies may involve considerable risk but may lead to a breakthrough in a particular area, or to the development of novel techniques, agents, methodologies, models, or applications that could have a major impact on a field of biomedical, behavioral, or clinical research.
Applications for R21 awards should describe projects distinct from those supported through the traditional R01 mechanism. For example, long-term projects, or projects designed to increase knowledge in a well-established area, will not be considered for R21 awards. Applications submitted under this mechanism should be exploratory and novel. These studies should break new ground or extend previous discoveries toward new directions or applications.

II. Award Information
1. Mechanism of Support
This FOA will use the NIH Exploratory/Developmental Research Grant (R21) award mechanism. As an applicant, you will be solely responsible for planning, directing, and executing the proposed project.
This FOA uses Just-in-Time information concepts. It also uses the modular as well as the non-modular budget formats (see the Modular Applications and Awards section of the NIH Grants Policy Statement. Specifically, if you are submitting an application with direct costs in each year of $250,000 or less (excluding consortium Facilities and Administrative [F&A] costs), use the PHS398 Modular Budget component provided in the SF424 (R&R) Application Package and SF424 (R&R) Application Guide (see specifically Section 5.4, Modular Budget Component, of the Application Guide).
All foreign applicants must complete and submit budget requests using the Research & Related Budget component found in the application package for this FOA. See NOT-OD-06-096, August 23, 2006.
Exploratory/Developmental grant support is for new projects only; competing renewal (formerly competing continuation ) or revised applications will not be accepted in response to this RFA. At this time, there are no plans to reissue this FOA.
2. Funds Available
Because the nature and scope of the proposed research will vary from
application to application, it is anticipated that the size and duration of
each award will also vary. Although the financial plans of the Institutes and
Centers (ICs) provide support for this program, awards pursuant to this funding
opportunity are contingent upon the availability of funds and the submission of
a sufficient number of meritorious applications.
The total project period for an application submitted in response to this funding opportunity may not exceed 2 years. Although the size of award may vary with the scope of research proposed, it is expected that applications will stay within the budgetary guidelines for an exploratory/developmental project; direct costs are limited to $275,000 over an R21 two-year period, with no more than $200,000 in direct costs allowed in any single year. Applicants may request direct costs in $25,000 modules, up to the total direct costs limitation of $275,000 for the combined two-year award period. NIH grants policies as described in the NIH Grants Policy Statement will apply to the applications submitted and awards made in response to this FOA.
The participating organization(s) NIMH intends to commit approximately $1,000,000 dollars in fiscal year 2008 to fund 3-4 applications in response to this RFA and its related RFA MH-08-110.

More
than one PD/PI, or multiple PDs/PIs, may be designated on the application for
projects that require a team science approach that clearly does not fit the
single-PD/PI model. Additional information on the implementation plans and policies and
procedures to formally allow more than one PD/PI on individual research

General
Characteristics of Responsive Applications
The goal of this initiative is to adopt circuit-based approaches to understanding the development and function of the prefrontal cortex in the context of other brain regions that regulate complex behaviors. To achieve this goal, proposed studies are required to incorporate techniques that go beyond simply correlating changes in neural activity in one or more regions with changes in behavior. In order to be judged responsive, applications submitted under this FOA must propose to use techniques that enable the precise temporal or spatial manipulation of specific circuits and must include a corresponding assessment of the effects of these manipulations on cognitive and/or emotional behavior. If an application is planning to use these techniques in rodents but not in non-human primates, a strong justification should be provided along with a clear plan for how these techniques can be adapted for use in non-human primates. Applications involving collaborations between molecular biologists developing genetic and molecular tools in rodents and neuroscientists examining the circuits that support complex behavior in non-human primates are strongly encouraged and will be deemed most relevant for this FOA. Preliminary data is not required for this FOA but can be included to demonstrate feasibility of the proposed approach. Applications proposing to use human subjects will not be accepted in response to this FOA.

Failure to include this data field will cause the application to be rejected.
All projects proposing Multiple PDs/PIs will be required to include a new section describing the leadership of the project.
Multiple PD/PI Leadership Plan: For applications designating multiple PDs/PIs, a new section of the research plan, entitled Multiple PD/PI Leadership Plan (Section 14 of the Research Plan Component in the SF424 (R&R)), must be included. A rationale for choosing a multiple PD/PI approach should be described. The governance and organizational structure of the leadership team and the research project should be described, including communication plans, process for making decisions on scientific direction, and procedures for resolving conflicts. The roles and administrative, technical, and scientific responsibilities for the project or program should be delineated for the PDs/PIs and other collaborators.
If budget allocation is planned, the distribution of resources to specific components of the project or the individual PDs/PIs should be delineated in the Leadership Plan. In the event of an award, the requested allocations may be reflected in a footnote on the Notice of Award.

Applications Involving Multiple Institutions
When multiple institutions are involved, one institution must be designated as the prime institution and funding for the other institution(s) must be requested via a subcontract to be administered by the prime institution. When submitting a detailed budget, the prime institution should submit its budget using the Research & Related Budget component. All other institutions should have their individual budgets attached separately to the Research & Related Subaward Budget Attachment(s) Form. 3.B. Submitting an Application Electronically to the NIH

5. Funding Restrictions
All NIH awards are subject to the terms and
conditions, cost principles, and other considerations described in the NIH Grants
Policy Statement.
Pre-award costs are allowable. A grantee may, at its
own risk and without NIH prior approval, incur obligations and expenditures to
cover costs up to 90 days before the beginning date of the initial budget
period of a new award if such costs: are necessary to conduct the project, and
would be allowable under the grant, if awarded, without NIH prior approval. If
specific expenditures would otherwise require prior approval, the grantee must
obtain NIH approval before incurring the cost. NIH prior approval is required
for any costs to be incurred more than 90 days before the beginning date of the
initial budget period of a new award.
The incurrence of pre-award costs in anticipation of a
competing or non-competing award imposes no obligation on NIH either to make
the award or to increase the amount of the approved budget if an award is made
for less than the amount anticipated and is inadequate to cover the pre-award
costs incurred. NIH expects the grantee to be fully aware that pre-award costs
result in borrowing against future support and that such borrowing must not
impair the grantee's ability to accomplish the project objectives in the
approved time frame or in any way adversely affect the conduct of the project.

Plan for Sharing Research Data
The precise content of the data-sharing plan will
vary, depending on the data being collected and how the investigator is
planning to share the data. Applicants who are planning to share data may wish
to describe briefly the expected schedule for data sharing, the format of the
final dataset, the documentation to be provided, whether or not any analytic
tools also will be provided, whether or not a data-sharing agreement will be
required and, if so, a brief description of such an agreement (including the
criteria for deciding who can receive the data and whether or not any
conditions will be placed on their use), and the mode of data sharing (e.g.,
under their own auspices by mailing a disk or posting data on their
institutional or personal website, through a data archive or enclave).
Investigators choosing to share under their own auspices may wish to enter into
a data-sharing agreement. References to data sharing may also be appropriate in
other sections of the application.
Applicants requesting more than $500,000 in direct costs in any year of
the proposed research must include a plan for sharing research data in their
application. The funding organization will be responsible for monitoring the

The reasonableness of the data sharing plan or the rationale for not sharing
research data may be assessed by the reviewers. However, reviewers will not

Investigators responding to this funding opportunity should include a sharing
research resources plan addressing how unique research resources will be shared
or explain why sharing is not possible.
The adequacy of the resources sharing plan and any
related data sharing plans will be considered by Program staff of the funding
organization when making recommendations about funding applications. The
effectiveness of the resource sharing will be evaluated as part of the

Applicants
should describe in detail how resources will be made available. For
mouse-related bioresources, it is strongly suggested that the applicant propose
to deposit mice into NIH-supported repositories in a
timely manner so that these lines can be made available to researchers and to

Applications submitted in response to this funding opportunity will compete for available funds with all other recommended applications. The following will be considered in making funding decisions:
The NIH R21 exploratory/developmental grant is a mechanism for supporting novel scientific ideas or new model systems, tools, or technologies that have the potential to significantly advance our knowledge or the status of health-related research. Because the Research Plan component is limited to 15 pages, an exploratory/developmental grant application need not have extensive background material or preliminary information as one might normally expect in an R01 application. Accordingly, reviewers will focus their evaluation on the conceptual framework, the level of innovation, and the potential to significantly advance our knowledge or understanding. Reviewers will place less emphasis on methodological details and certain indicators traditionally used in evaluating the scientific merit of R01 applications, including supportive preliminary data. Appropriate justification for the proposed work can be provided through literature citations, data from other sources, or, when available, from investigator-generated data. Preliminary data are not required for R21 applications; however, they may be included if available.
The goals of NIH supported research are to advance our understanding of biological systems, to improve the control of disease, and to enhance health. In their written critiques, reviewers will be asked to comment on each of the following criteria in order to judge the likelihood that the proposed research will have a substantial impact on the pursuit of these goals. Each of these criteria will be addressed and considered in assigning the overall score, weighting them as appropriate for each application.
Note that an application does not need to be strong in
all categories to be judged likely to have major scientific impact and thus
deserve a high priority score.
Significance: Does this study address an important problem? If the aims of the application are
achieved, how will scientific knowledge or clinical practice be advanced? What
will be the effect of these studies on the concepts, methods, technologies,
treatments, services, or preventative interventions that drive this field?
Approach: Are the conceptual or clinical framework, design, methods, and
analyses adequately developed, well integrated, well reasoned, and appropriate
to the aims of the project? Does the applicant acknowledge potential problem
areas and consider alternative tactics?
Innovation: Is the project original and innovative? For example: Does
the project challenge existing paradigms or clinical practice; address an
innovative hypothesis or critical barrier to progress in the field? Does the
project develop or employ novel concepts, approaches, methodologies, tools, or
technologies for this area?
Investigators: Are the PD/PIs appropriately trained and well suited to
carry out this work? Is the work proposed appropriate to the experience level
of the principal investigator and other researchers? Does the investigative
team bring complementary and integrated expertise to the project (if
applicable)?
Environment: Does the scientific environment in which the work will be done contribute to the probability of success? Do the proposed studies benefit from unique features of the scientific environment, or subject populations, or employ useful collaborative arrangements? Is there evidence of institutional support?
Special Review Criteria: Does the research proposed use state-of-the-art techniques that enable the temporal or spatial manipulation of PFC-related circuits to examine the how the PFC causally regulates cognitive and/or emotional behavior? If the research includes the use of rodents, is there a strong justification that the experiments will provide significantly new insights into how the PFC regulates complex mental function and would the approach be adaptable for near-term use in non-human primates?
2.A. Additional Review Criteria
In addition to the above criteria, the following items
will continue to be considered in the determination of scientific merit and the
priority score:
Care and Use of Vertebrate Animals in Research: If vertebrate animals
are to be used in the project, the five items described under item 11 of the Research
Plan component of the SF424 (R&R) will be assessed.
Biohazards: If materials or procedures are proposed that are potentially
hazardous to research personnel and/or the environment, determine if the
proposed protection is adequate.
2.B. Additional Review Considerations
Budget: The reasonableness of the proposed budget and the requested
period of support in relation to the proposed research. The priority score
should not be affected by the evaluation of the budget.
Applications from Foreign Organizations: Whether the project presents special opportunities for furthering research programs through the use of unusual talent, resources, populations, or environmental conditions in other countries that are not readily available in the United States or that augment existing U.S. resources will be assessed

the organism, the timeline, the applicant's decision to distribute the resource
or deposit it in a repository, and other relevant considerations. For the R21
mechanism, the presence or adequacy of a plan should not enter into the scoring
of the application.

1. Award Notices
After the peer review of the application is completed, the PD/PI will be able
to access his/her Summary Statement (written critique) via the NIH eRA Commons.
If the application is under consideration for funding,

A formal notification in the form of a Notice of Award
(NoA) will be provided to the applicant organization. The NoA signed by the
grants management officer is the authorizing document. Once all administrative
and programmatic issues have been resolved, the NoA will be generated via email
notification from the awarding component to the grantee business official.
Selection of an application for award is not an
authorization to begin performance. Any costs incurred before receipt of the
NoA are at the recipient's risk. These costs may be reimbursed only to the

Human Subjects Protection:

A-110 has been revised to provide access to research data through the Freedom

action that has the force and effect of law (i.e., a regulation) may be

Applicants may wish to place data collected under this funding opportunity in a
public archive, which can provide protections for the data and manage the

At the same time the NIH recognizes the rights of grantees and contractors to
elect and retain title to subject inventions developed with Federal funding
pursuant to the Bayh Dole Act (see the NIH
Grants Policy Statement). Beginning October 1, 2004, all investigators
submitting an NIH application or contract proposal are expected to include in
the application/proposal a description of a specific plan for sharing and
distributing unique model organism research resources generated using NIH
funding or state why such sharing is restricted or not possible. This will
permit other researchers to benefit from the resources developed with public
funding. The inclusion of a model organism sharing plan is not subject to a
cost threshold in any year and is expected to be included in all applications
where the development of model organisms is anticipated.
Inclusion of Women And Minorities in Clinical
Research:
It is the policy of the NIH that women and members of
minority groups and their sub-populations must be included in all NIH-supported
clinical research projects unless a clear and compelling justification is provided
indicating that inclusion is inappropriate with respect to the health of the
subjects or the purpose of the research. This policy results from the NIH
Revitalization Act of 1993 (Section 492B of Public Law 103-43). All
investigators proposing clinical research should read the "NIH Guidelines

The amended policy incorporates: the use of an NIH definition of clinical

issued final modification to the "Standards for Privacy of Individually
Identifiable Health Information", the "Privacy Rule", on August

The PHS strongly encourages all grant recipients to
provide a smoke-free workplace and discourage the use of all tobacco products.
In addition, Public Law 103-227, the Pro-Children Act of 1994, prohibits
smoking in certain facilities (or in some cases, any portion of a facility) in
which regular or routine education, library, day care, health care, or early
childhood development services are provided to children. This is consistent

Loan Repayment Programs:
