Research Objectives
INTRODUCTION
Dietary habits are recognized as an important modifiable environmental factor influencing cancer risk and tumor behavior. Estimates suggest that 30-70% of all cancer cases might be preventable by diet, depending on the dietary components and specific type of cancer. Epidemiologic evidence indicates, for instance, that regular consumption of fruits, vegetables and whole grains may reduce cancer risk in some individuals. This association has been attributed to numerous bioactive compounds present in these foods, including essential nutrients, polyunsaturated fatty acids and many phytochemicals. An attractive possibility emerges that because of synergistic interactions among various bioactive components, whole foods (either as single food products or food combinations) may be more efficacious in terms of cancer prevention than isolated individual compounds.
BACKGROUND
Specific food types and dietary factors are increasingly implicated in health benefits. For example, combinations of wine, fish, dark chocolate, fruits, vegetables, garlic, and almonds have been suggested to offer special additive benefits in cardiovascular disease risk. Whether or not these, or possibly other, food combinations offer discernible protection against cancer and/or modify tumor behavior remains to be rigorously evaluated. Nonetheless, some studies do suggest that combinations of foods may offer special advantages for cancer prevention. For example, soy phytochemicals combined with green tea extracts have been reported to be more effective in inhibiting tumor angiogenesis, reducing estrogen receptor (ER)-alpha and lowering serum insulin-like growth factor (IGF)-1 than the same substances administered alone. Clearly, a better understanding is needed of the bioactive components present in food as well as the mechanism(s) by which these dietary components may contribute towards cancer prevention.
Malignant cells are characterized by the upregulation of multiple signaling pathways that promote proliferation, inhibit apoptosis, promote angiogenesis in the case of solid tumors, and enable cancer cells to invade and migrate through tissues. Down regulation or loss of proteins and pathways that oppose these behaviors is also commonly seen. During the carcinogenic process, cells tend to acquire multiple oncogenic mutations that are often functionally redundant. Consequently, inhibiting any single dysregulated pathway may have only a modest impact on tumor behavior.
Targeting multiple molecular targets that are prone to dysregulation in a given type of cancer seems to offer advantages for cancer prevention. Accordingly, various bioactive food components have the potential to affect a multitude of cancer-relevant process, such as carcinogen metabolism, DNA repair, cell proliferation, apoptosis, differentiation, and angiogenesis. Whereas the ability of dietary components to modify multiple steps in the cancer process simultaneously is desirable, it complicates the assessment of cancer cell responses. Also, some targets can be more important for specific dietary components or their combinations than other targets. Thus, it is essential to establish which targets/processes are most relevant for the reduction of tumor incidence and/or modification of tumor behavior. Furthermore, since many of the potentially targetable processes are likely influenced by several food components, it is necessary to obtain a better understanding of physiologically relevant synergistic and antagonistic interactions among food components.
Individual foods versus their isolated constituents
There is evidence that intake of bioactive dietary components in the form of various foods may offer advantages over the intake of isolated constituents. Factors present in the foods may influence the absorption, metabolism, and/or retention of the bioactive food components. In addition, multiple as yet unidentified bioactive compounds may be present within the food. For example, consumption of tomato products may be more beneficial for prostate cancer prevention than the consumption of isolated lycopene, a presumed major cancer risk reducing component in tomato products. Fat-soluble vegetable extracts may be more efficacious than carotene in inhibiting cell proliferation and inducing morphologic changes consistent with apoptosis (cellular shrinkage, chromatin condensation, and nuclear fragmentation) in cultured cancer cells. However, in other cases the food may not be as effective as its isolated components, suggesting the food may contain constituents that inhibit a desirable cellular response. Such inhibitory effects may again reflect modifying the absorption and/or metabolism of cancer-preventing bioactive food constituent(s) or by counteracting their action(s) on cellular targets. Such antagonistic components may explain, for instance, the reduced ability of soy flour and full-fat soy flakes (whole soybean) to inhibit aberrant crypt foci compared to isolated genistein. A better understanding is needed of how the bioactive components either in isolation or as part of a food matrix interact at the cellular and molecular levels to optimize diet-based strategies for reducing cancer risks.
Combinations of cancer protective components
Strategies that use a combination of agents with distinct molecular mechanisms, rather than individual agents, offer a potential for maximizing cancer prevention while minimizing toxicity. This principle, which is routinely applied to therapeutic treatments, is fully applicable to cancer-prevention by food components. Moreover, as shown in recent studies, dietary components have the potential to enhance the cancer preventive effects of pharmaceutical agents by modifying cancer-relevant molecular targets. For example, diets containing high levels of olive oil exert a significant protective effect against colon tumor development that enhances the inhibitory effects of an anticancer drug sulindac. This enhancement is likely related to the observed downregulation of the expression and activity of key proteins involved in prostaglandin-biosynthesis (COX-2) and apoptosis-induction (Bcl-2 and caspase-3) pathways. Another example for a desirable interaction between a food component and a pharmaceutical agent includes the ability of the soy isoflavone daidzen to improve the capacity of tamoxifen, to prevent mammary tumors.
Dietary components may affect the same molecular targets as drugs used for cancer prevention. Such overlap may allow lower amounts of the drug to be utilized and thus reduce potential adverse effects of the drug. For example, a diet enriched in omega-3 fatty acids can lower COX-2 activity and, thereby, exert synergistic effects with low doses of the COX-2 inhibitor celecoxib for colon cancer prevention. Similarly, the food components docosohexaenoic acid (DHA) and genistein attenuate the induction of HMG-CoA reductase activity in mevastatin-treated MCF-7 cells. These observations suggest that dietary genistein or DHA may lower the dose of statin required to achieve the same level of HMG-CoA reductase inhibition, thereby enhancing the effects of statins in breast cancer prevention. Finally, the combination of vitamin D3 and genistein causes growth inhibition of prostate cancer cells at biologically achievable concentrations (that for both compounds are lower than respective growth inhibitory concentrations of single agents). Genistein appears to potentiate the growth inhibitory and pro-apoptotic action of vitamin D3 by directly inhibiting CYP24 (25-hydroxyvitamin D(3)-24-hydroxylase) enzyme activity. This inhibition increases the half-life of vitamin D3, which, in turn, results in homologous up-regulation of the cellular vitamin D receptor. The mentioned examples point to the advantages of combining pharmacological agents and bioactive food components. Unfortunately, however, analogous knowledge regarding combinations of various bioactive food components remains limited.
Dietary components that alter multiple molecular targets within a specific biological process or pathway have been shown to have a potential for synergistic effects. For example, quercetin and genistein synergistically inhibit proliferation of ovarian carcinoma cells by modifying different stages in the cell cycle and different signal transduction pathways. Quercetin arrests the cell cycle at the G1 and’s phase boundary whereas genistein affects the G2 and/or early M phase. A selenium compound and vitamin E have been shown to have synergistic effects on apoptosis induction in human prostate cancer cells. The observed apoptosis enhancement might reflect different patterns of caspase activation produced by the selenium compound and vitamin E. These examples illustrate the point that combinations of dietary components with different mechanisms of action may have synergistic effects in cancer prevention.
An important understudied area is whether or not additive effects may be expected for different dietary components that modulate the same molecular target. For example, both garlic organosulfur compounds and sulforaphane (which is present in broccoli) induce expression of detoxifying enzymes mediated by the binding of the transcription factor Nrf2 to the antioxidant response elements (ARE) located in the promoter regions of Nrf2-regulated genes. Will this common target become saturated if an individual consumes high levels of garlic organosulfur compounds preventing broccoli-derived sulforaphane from exerting any beneficial effects? However, the mentioned dietary components also influence other targets, in addition to Nrf2-ARE binding. Do these other targets become more important for bringing about a phenotypic change?
RESEARCH OBJECTIVES AND SCOPE
This Funding Opportunity Announcement (FOA) is designed to stimulate investigations that would characterize interrelationships among food components and/or food combinations. The emphasis is on targeting multiple sites in the cancer process to maximize cancer prevention and minimize potential toxicity. In addition, the proposed investigations must concentrate on those effects of dietary bioactive components that are relevant to the levels of these substances achievable via normal dietary or supplement intake.
Proposed projects should apply new high-throughput genomic, epigenomic, proteomic and metabolomic technologies to either evaluate multiple molecular targets within a cancer process or multiple processes. Priority should be on targets and processes that have already been implicated in the effects of given dietary components but target discovery research is also eligible. The main outcome of these studies must be the prioritization of targets/processes that are most relevant to cancer prevention by dietary components. The overall long-term goal is to use such information in developing dietary strategies to reduce cancer risk and/or modify tumor behavior.

Opportunity Announcement (FOA) will use the NIH Research Project Grant (R01) award mechanism.
The applicant will be solely responsible for planning, directing, and executing the proposed project.
This FOA uses Just-in-Time information concepts. It also uses the modular as well as the non-modular budget formats (see http://grants.nih.gov/grants/funding/modular/modular.htm).
Specifically, if you are a United States (U.S.) organization and are submitting an application with direct costs in each year of $250,000 or less (excluding consortium Facilities and Administrative [F&A] costs), use the PHS398 Modular Budget component provided in the SF424 (R&R) Application Package and SF424 (R&R) Application Guide (see specifically Section 5.4, Modular Budget Component, of the Application Guide).

of the proposed research will vary from application to application, it is
anticipated that the size and duration of each award will also vary. Although
the financial plans of the NCI provide support for this program, awards
pursuant to this funding opportunity are contingent upon the availability of
funds and the submission of a sufficient number of meritorious applications.

Individuals from underrepresented racial and ethnic groups as well as individuals with disabilities are always encouraged to apply for NIH support.
More than one PD/PI, or multiple PDs/PIs, may be designated on the application for projects that require a team science approach that clearly does not fit the single-PD/PI model. All other PDs/PIs should be listed in the Research & Related Senior/Key Person component and assigned the project role of PD/PI. Please remember that all PDs/PIs must be registered in the eRA Commons prior to application submission. The Commons ID of each PD/PI must be included in the Credential field of the Research & Related Senior/Key Person component. Failure to include this data field will cause the application to be rejected.
All projects proposing Multiple PDs/PIs will be required to include a new section describing the leadership of the project.
Multiple PD/PI Leadership Plan: For applications designating multiple PDs/PIs, a new section of the research plan, entitled Multiple PD/PI Leadership Plan (Section 14 of the Research Plan Component in the SF424 (R&R)), must be included. A rationale for choosing a multiple PD/PI approach should be described. The governance and organizational structure of the leadership team and the research project should be described, including communication plans, process for making decisions on scientific direction, and procedures for resolving conflicts. The roles and administrative, technical, and scientific responsibilities for the project or program should be delineated for the PDs/PIs and other collaborators.
If budget allocation is planned, the distribution of resources to specific components of the project or the individual PDs/PIs should be delineated in the Leadership Plan. In the event of an award, the requested allocations may be reflected in a footnote on the Notice of Award.
Applications Involving a Single Institution

Applications Involving Multiple Institutions
When multiple institutions are involved, one institution must be designated as the prime institution and funding for the other institution(s) must be requested via a subcontract to be administered by the prime institution. When submitting a detailed budget, the prime institution should submit its budget using the Research & Related Budget component. All other institutions should have their individual budgets attached separately to the Research & Related Subaward Budget Attachment(s) Form. All NIH awards are subject to the terms and conditions, cost principles, and other considerations described in the NIH Grants Policy Statement.
Pre-award costs are
allowable. A grantee may, at its own risk and without NIH prior approval, incur
obligations and expenditures to cover costs up to 90 days before the beginning
date of the initial budget period of a new or competing renewal (formerly
competing continuation ) award if such costs: are necessary to conduct the
project, and would be allowable under the grant, if awarded, without NIH prior
approval. If specific expenditures would otherwise require prior approval, the
grantee must obtain NIH approval before incurring the cost. NIH prior approval
is required for any costs to be incurred more than 90 days before the beginning
date of the initial budget period of a new or competing renewal award.
The incurrence of pre-award costs in
anticipation of a competing or non-competing award imposes no obligation on NIH
either to make the award or to increase the amount of the approved budget if an
award is made for less than the amount anticipated and is inadequate to cover
the pre-award costs incurred. NIH expects the grantee to be fully aware that
pre-award costs result in borrowing against future support and that such
borrowing must not impair the grantee's ability to accomplish the project
objectives in the approved time frame or in any way adversely affect the

The applicant organization must include its DUNS number in its Organization Profile in the eRA Commons. This DUNS number must match the DUNS number provided at CCR registration with Grants.gov. Items 2-5 of the PHS398 Research Plan component are limited to 25 pages. requesting $500,000 or more in direct costs for any year (excluding consortium

you are developing plans for the study;

Obtain agreement from the IC staff that the IC will accept your application for
consideration for award; and,
3)

IMPORTANT NOTE: NIH has published new limitations on grant application appendix materials to encourage applications to be as concise as possible while containing the information needed for expert scientific review.

Applicants who are planning to share data may wish to describe briefly the expected schedule for data sharing, the format of the final dataset, the documentation to be provided, whether or not any analytic tools also will be provided, whether or not a data-sharing agreement will be required and, if so, a brief description of such an agreement (including the criteria for deciding who can receive the data and whether or not any conditions will be placed on their use), and the mode of data sharing (e.g., under their own auspices by mailing a disk or posting data on their institutional or personal Web site, through a data archive or enclave). Investigators choosing to share under their own auspices may wish to enter into a data-sharing agreement. References to data sharing may also be appropriate in other sections of the application.
Experimental data for sharing, their format, analytical algorithms, computational modeling and visualizations, and other bioinformatics tools resulting from this FOA must be compatible with the NIH-approved bioinformatics platforms, such as those designed and implemented by the NCI Center for Bioinformatics (http://ncicb.nci.nih.gov), ultimately including caBIG, an open source/open access information network for the sharing of cancer research data and related software tools (https://cabig.nci.nih.gov).

Approach: Are the conceptual or clinical framework, design, methods,
and analyses adequately developed, well-integrated, well-reasoned, and
appropriate to the aims of the project? Does the applicant acknowledge
potential problem areas and consider alternative tactics? For
applications designating multiple PIs, is the leadership approach, including the designated
roles and responsibilities, governance, and organizational structure,
consistent with and justified by the aims of the project and the expertise of
each of the PDs/PIs? Are the administrative
plans for the management of the research project appropriate, including plans
for resolving conflicts?
Innovation: Is the project original and innovative? For example: Does the project challenge existing paradigms or clinical practice; address an innovative hypothesis or critical barrier to progress in the field? Does the project develop or employ novel concepts, approaches or methodologies, tools, or technologies for this area?
Investigators: Are the PD/PIs and other key personnel appropriately
trained and well suited to carry out this work? Is the work proposed
appropriate to the experience level(s) of the principal investigator(s) and
other researchers? Do the PD/PI(s) and investigative
team bring complementary and integrated expertise to the project (if applicable)?
Environment: Do(es) the scientific environment(s) in which the work will
be done contribute to the probability of success? Do the proposed studies
benefit from unique features of the scientific environment(s), or subject

Protection of Human Subjects from Research Risk: The involvement of human
subjects and protections from research risk relating to their participation in

Inclusion of Women, Minorities and Children
in Research: The adequacy of plans to include subjects from both

potentially hazardous to research personnel and/or the environment, determine

Recipients of PHS support for activities involving

Model Organisms:
NIH is committed
to support efforts that encourage sharing of important research resources including
the sharing of model organisms for biomedical research (see http://grants.nih.gov/grants/policy/model_organism/index.htm).
At the same time, the NIH recognizes the rights of grantees and contractors to
elect and retain title to subject inventions developed with Federal funding
pursuant to the Bayh-Dole Act (see the NIH
Grants Policy Statement. Beginning October 1, 2004, all investigators
submitting an NIH application or contract proposal are expected to include in
the application/proposal a description of a specific plan for sharing and
distributing unique model organism research resources generated using NIH
funding or state why such sharing is restricted or not possible. This will

Inclusion of
Women And Minorities in Clinical Research:
It is the policy
of the NIH that women and members of minority groups and their sub-populations
must be included in all NIH-supported clinical research projects unless a clear
and compelling justification is provided indicating that inclusion is
inappropriate with respect to the health of the subjects or the purpose of the
research. This policy results from the NIH Revitalization Act of 1993 (Section
492B of Public Law 103-43). All investigators proposing clinical research
should read the "NIH Guidelines for Inclusion of Women and Minorities as

2010:
The Public Health
Service (PHS) is committed to achieving the health promotion and disease
prevention objectives of "Healthy People 2010," a PHS-led national
activity for setting priority areas. This PA is related to one or more of the
priority areas. Potential applicants may obtain a copy of "Healthy People

Authority and
Regulations:
This program is described in
the Catalog of Federal Domestic Assistance at http://www.cfda.gov/ and is not subject to the
intergovernmental review requirements of Executive
Order 12372 or Health Systems Agency review. Awards are made under the
authorization of Sections 301 and 405 of the Public Health Service Act as
amended (42 USC 241 and 284) and under Federal Regulations 42 CFR Part 52 and 45 CFR Parts 74 and 92. All awards are subject to the
terms and conditions, cost principles, and other considerations described in
the NIH Grants Policy Statement.
The PHS strongly encourages
all grant recipients to provide a smoke-free workplace and discourage the use
of all tobacco products. In addition, Public Law 103-227, the Pro-Children Act
of 1994, prohibits smoking in certain facilities (or in some cases, any portion
of a facility) in which regular or routine education, library, day care, health
care, or early childhood development services are provided to children. This is
consistent with the PHS mission to protect and advance the physical and mental
health of the American people.
Loan Repayment
Programs:
NIH encourages
applications for educational loan repayment from qualified health professionals
who have made a commitment to pursue a research career involving clinical,
pediatric, contraception, infertility, and health disparities related areas.
The LRP is an important component of NIH's efforts to recruit and retain the
next generation of researchers by providing the means for developing a research
career unfettered by the burden of student loan debt. Note that an NIH grant is
not required for eligibility and concurrent career award and LRP applications
are encouraged. The periods of career award and LRP award may overlap providing
the LRP recipient with the required commitment of time and effort, as LRP
awardees must commit at least 50% of their time (at least 20 hours per week
based on a 40 hour week) for 2 years to the research. For further information,
